摘要
目的建立一种高效液相色谱法(HPLC)检测大鼠血浆中西咪替丁的浓度。方法采用BDS HYPERSIL C18(5μm,4.6×250mm)色谱柱,流动相为甲醇-水-二乙胺-磷酸(25:75:0.47:0.38);流速:1m L·min-1;检测波长:225nm;进样量20μL;柱温30℃。结果西咪替丁在1μg·mL^(-1)~100μg·mL^(-1)浓度范围内与峰面积呈良好线性关系(r=0.9999);样品平均回收率为94%~104%,日内、日间RSD均低于5%(n=5)。结论本法专属性好,灵敏度高,操作简便,可用于大鼠血浆中西咪替丁含量的测定。
OBJECTIVE To establish an HPLC method for determination of cimetidine in rat plas- ma. METHODS The sample was analyzed in column BDS HYPERSIL C 18 (5μm,4. 6 × 250mm) ;mobile phase consisted of methonal-water-diethylamine-phosphate(25:75:0. 47: 0. 38 ) ;The flow rate was 1.0mL ·min-1;The de- tective wavelength was 225nm. The sample size was 20μL. The detection temperature was at 30℃. RESULTS The calibration curves of cimetidine showed good linerarity ,in the range of 11μg · mL-1 - 100μg · mL-l ( r = 0. 9999 ) ; The average recoveries of sample were in the range of 94% -104%. the intra-day and inter-day variation(RSD) was less than 5% ( n = 5 ). CONCLUSION This method is accurate, simple, quick and with good sensitivity for determi- ning the cimetidine concentration in rat plasma.
