健脾补土法组方对脑缺血/再灌注大鼠脑组织INTβ_3、ILK及FAK mRNA及蛋白表达的影响 被引量:6

The Effect of Spleen-strengthening Therapy on the mRNA and protein expression levels of INTβ_3,ILK and FAK in Rats with Cerebral Ischemia/Reperfusion Injury

摘要 目的研究健脾补土法组方对脑缺血/再灌注损伤大鼠测定整合素β3(INTβ_3)、ILK、FAK mRNA及蛋白的表达影响。方法将36只雄性SD大鼠随机分为假手术组、模型组、依达拉奉组、健脾补土方小剂量组、健脾补土方中剂量组、健脾补土方大剂量组6小组,每组6只。采用线栓法制备MCAO模型,7天后处死,应用RT-PCR法测定缺血再灌注后缺血区脑皮质内INTβ_3、ILK、FAK mRNA的表达的含量变化及运用健脾补土法组方对其含量的影响,Western blot法检测其蛋白含量变化。结果模型组与假手术组相比,模型组INTβ_3、ILK、FAK蛋白以及mRNA含量显著减少(P<0.01);与模型组比较,依达拉奉组、健脾补土小剂量、健脾补土中剂量组、健脾补土大剂量组整合素β3、ILK、FAK蛋白以及mRNA含量增加,差异有统计学意义(P<0.05或P<0.01)。结论健脾补土法组方脑保护作用可能与促进INTβ_3、ILK、FAK表达有关。 Objective To study the effect of spleen-strengthening therapy on the expression of INT,ILK and FAK in rats with cerebral ischemia/reperfusion injury.Methods 36 male SD rats were randomly divided into sham-operation group,modelling group,edaravone group and,low-dosed spleen-strengthening group,middle-dosed spleen-strengthening group,high-dosed spleen-strengthening group,6 in each group.The model of MCAO was induced by middle cerebral artery occlusion and recanalization,and execute them after treated 7 days.RT-PCR was used to detect the mRNA expression levels of INT,ILK and FAK.Western blot was used to detect the protein expression levels of INTβ_3,ILK and FAK.Results Compared with the modelling group,the mRNA and protein expression levels of INTβ_3,ILK and FAK was obviously decreased(P0.01).Compared with the modelling group,the mRNA and protein expression levels of INT,ILK and FAK was obviously increased(P0.05,P0.01) in low-dosed spleen-strengthening group,middle-dosed spleen-strengthening group and high-dosed spleen-strengthening group.Conclusion The cerbral protection effect of prescription of spleen-strengthening may related to its promoting the expression of INTβ_3,ILK and FAK.
