黄芩糖蛋白提取工艺的正交实验优选 被引量:2

摘要 目的建立黄芩糖蛋白的最佳提取工艺。方法从提取黄芩的pH值、温度、液料比、盐浓度、提取时间5个方面进行实验确定黄芩糖蛋白的最佳提取工艺。选取pH值、温度、液料比做正交试验,对正交试验结果进行方差分析和重复验证。采用Tris-HCl缓冲液提取法,考马斯亮蓝显色法测定黄芩糖蛋白含量,计算出糖蛋白得率。通过比较各条件下黄芩糖蛋白得率,确定黄芩糖蛋白提取的最佳条件。结果用pH=8、液料比为1:10、盐浓度0.15 mol/L的Tris-HCl缓冲液在20℃下水浴提取1.5 h,黄芩糖蛋白得率为2.94%。结论该实验首次确定黄芩糖蛋白提取最佳条件,为其糖蛋白的分离纯化与药理活性的研究提供研究基础。 Objective To establish the best extraction technology of Radix Scutellariae glycoprotein.Methods From Radix Scutellariaeicalensis extract liquid of pH,temperature,liquid ratio,salt concentration,extracting time five aspects carries on the experiment to determine the best extraction technology of Radix Scutellariae glycoprotein.The effects of pH,temperature,liquid ratio,on the extraction amount were analyzed by the orthogonal experiment and repeated the validation test.The Tris-HCl buffer extraction and the method of coomassie brilliant blue color determine the concentration of sugar and protein,and then calculate the glycoprotein yield,and then by comparing the Radix Scutellariae glycoprotein under the condition of yield,to determine the best condition of Radix Scutellariae glycoprotein extraction.Results The 1:10 buffer of pH=8,liquid feed ratio and salt concentration of Tris-HCl were extracted at 20 for 1.5 hours,and the highest yield of glycoprotein was 2.94%.Conclusion The experiment was the first to determine the process of optimal glycoprotein extraction from Radix Scutellariae and provided the basis for the purification and pharmacological activities of the glycoprotein.
