脂肪源性神经干细胞细胞移植对帕金森病大鼠脑内氧化应激损伤的影响 被引量:5

摘要 目的探讨脂肪源来源的神经干细胞细胞移植对帕金森病(Parkinson,s disease,PD)大鼠模型脑内氧化应激损伤的影响。方法获取SD大鼠脂肪干细胞,利用神经营养因子将其诱导为神经干细胞。用6羟基多巴胺(6-OHDA)大鼠纹状体内两点毁损法制作PD模型(旋转圈数>7 r/min),将成功大鼠模型随机分为模型组和神经干细胞移植组,每组15只,另选15只正常大鼠为对照组。纹状体区立体定位细胞移植后8周,进行模型组和细胞移植组大鼠旋转圈数的行为学检测。化学比色法分别检测PD模型组、神经干细胞移植组、正常对照组大鼠脑内超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-PX)活性以及丙二醛(MDA)含量。免疫组化法计数大鼠脑内酪氨酸羟化酶(TH)数量。结果细胞移植后8周,细胞移植组40 min内大鼠旋转圈数(19±4)较PD模型组旋转圈数(38±3)减少,大鼠脑内SOD、GSH、GSH-Px活性提高,MDA含量减少。TH计数表明,细胞移植组(22.40±3.89)、模型组(13.50±1.08)及正常对照组(43.20±3.32)三组比较差异有统计学意义(P<0.05)。结论脂肪来源的神经干细胞移植治疗帕金森病模型大鼠可改善其行为活动,其机制可能与减轻大鼠脑内6-OHDA所致的氧化应激损伤有关。 Objective To explore the effects of adipose-derived neural stem cell transplantation on oxidative stress reaction in rats with Parkinson' s disease (PD). Methods The adipose stem cells from SD rat fat tissues were isolated and induced to develop the neural stem cell with neurotrophic factors. Meanwhile, the PD model was established through brain stereotoxic injection of 6-OHDA (7 r/rain). The PD model rats were randomly divided into two groups:model group (n = 15 ) and neural stem cell transplantation group (n = 15 ). Fifteen normal SD rats were chosen as normal group. The behavior( number of rotation) was observed in model group and cell transplan- tation group at 8 week. Moreover, the contents of superoxidedismutase(SOD) and glutathione (GSH), the activity of glutathione peroxi- dase( GSH - Px) and malondialdehyde(MDA) in rat brain nigrostriatum were detected by chemical colorimetry. The number of TH in striatum was detected by immunohistochemical staining. Results At 8 week, the rotation in 40 min was lower in neural stem cell transplantation group than in model group (19±4 vs 38±3, P 〈 0. 05). The contents of SOD, GSH, GSH-PX were increased compared with PD model group and normal group, while the content of MDA was decreased significantly. The quantity of TH were 22. 40 ±3. 89 in cell transplantation group, 13.50 ±1.08 in model group, and 43.20 ± 3.32 in normal group, and the difference was statistically significant(P 〈 0. 05). Conclusion The adipose-derived neural stem cell transplantation can improve the behavior of PD rats, which may be related to the decrease of the injury of oxidative stress.
