摘要
为了研究姜黄素对葡聚糖硫酸钠(DSS)诱导的血管内皮细胞损伤的保护作用,试验采用DSS建立血管内皮细胞损伤模型,采用流式细胞术分析线粒体膜电位(MMP)和细胞凋亡情况;MTT法检测细胞增殖抑制率。结果表明:随DSS浓度的增加,细胞增殖受到抑制,MMP下降,细胞凋亡增加;姜黄素提高了细胞MMP,降低了细胞凋亡百分率和增殖抑制率。说明姜黄素通过抑制MMP的下降而减轻了DSS诱导血管内皮细胞的凋亡。
To study the protective effect of cureumin on the dextran sulfate sodium (DSS) - induced injury of vascular endothelial cells, DSS was used to establish an injury model of vascular endothelial ceils. The mitochondfial membrane potential (MMP) and apoptosis were analyzed by flow eytometry. MTT assay was used to detect the inhibition rate of cell proliferation. The results showed that the increase in DSS concentra- tion the cell proliferation was inhibited with the increase of DSS concentration, while the MMP was decreased and the apoptosis was increased. Curcumin increased the MMP of cells and decreased the percentage of apoptosis and the proliferation inhibition rate. The results indicate that curcumin can relieve the apoptosis of vascular endothelial cells induced by DSS through inhibiting the decrease of MMP.
