摘要
目的通过对东方拟无枝酸菌的调控基因进行研究及改造以获得高产万古霉素的突变株。方法选取经基因组测序分析获得的6个推测具有调控作用的基因:strR型的AORI_1475、araC型的AORI_1832、AORI_2956、AORI_3531和lysR型的AORI_3398、AORI_7412进行过表达实验。结果发现AORI_1475、AORI_1832、AORI_2956、AORI_3531的过表达分别将万古霉素产量上调了55%、51%、18%、29%,而AORI_3398、AORI_7412却使万古霉素产量下降了12%左右。结论初步判断AORI_1475、AORI_1832、AORI_2956、AORI_3531对万古霉素合成有正调控作用,而AORI_3398、AORI_7412对万古霉素合成起负调控作用。
Objective The objective of this study was to obtain the mutant strains with high productivity of vancomycin through studying and transformation of regulatory genes inAmycolatopsisorientalis. Methods Six putative regulatory genes AORI1475, AORI1832, AORI2956, AORI3531, AORI3398 and AORI7412 were selected based on DNA sequence analysis of HCCB10007 genome, and overexpressed in this study, respectively. Results The yields of vancomycin of the first four overexpressed strains were increased by 55%, 51%, 18%, 29% while the last two were reduced by about 12%. Conclusion AORI1475, AORI1832, AORI2956 or AORI3531 plays positively regulatory roles in vancomycin synthesis while AORI3398 and AORI7412 negatively regulate the synthesis. Furthermore, the regulatory role of AORI2956, AORI3531, AORI3398 or AORI7412 is weak.
