枸橼酸铁铵对BV2小胶质细胞DMT1和FPN1表达影响 被引量:1

摘要 目的观察枸橼酸铁铵(FAC)对BV2小胶质细胞二价金属离子转运蛋白1(DMT1)和铁转出蛋白1(FPN1)表达的影响。方法以100μmol/L的FAC处理BV2小胶质细胞24h,采用Western Blot方法观察细胞内DMT1和FPN1蛋白表达的变化。结果 FAC处理BV2小胶质细胞24h后,FAC组DMT1蛋白表达与对照组比较明显下调(t=3.880,P<0.01),但FPN1蛋白的表达明显上调(t=4.409,P<0.01)。结论 FAC处理BV2小胶质细胞24h,可使细胞中DMT1蛋白表达下调,FPN1蛋白表达上调,此过程可能与铁调节蛋白的调控有关。 Objective To investigate the impact of ferric ammonium citrate(FAC)on the expressions of divalent metal transporter 1(DMT1) and ferroportin 1(FPN1)in BV2 cells. Methods BV2 microglia cells were treated with 100μmol/L FAC for 24 h.Using Western Blot,the changes of DMT1 and FPN1 protein expression in BV2 microglia cells were observed. Results After 24 htreatment with FAC,the expression of DMT1 in BV2 microglia cells was lower than that in the control group(t=3.880,P〈0.01),and that of FPN1 was higher(t=4.409,P〈0.01). Conclusion After treatment with FAC for 24 h,the expression of DMT1 protein in BV2 microglia cells was down regulated,and that of FPN1 up regulated,this process may be related to the regulation of iron regulatory protein.
