摘要
目的通过观察小鼠灌胃给药后的中毒反应,考察益肾通络方浓缩合剂的安全性。方法将益肾通络方浓缩合剂以最大浓度(2.5 g/ml)和最大体积(0.4 ml/10 g)24 h内3次给药,连续观察14d内小鼠的活动情况和死亡数量,小鼠死亡0只,未测出LD50,测其最大耐受量。结果给药后14 d内小鼠生存状态良好,活动和饮食情况均无变化,部分小鼠有轻微腹泻,无死亡,空白组与给药组体重无统计学明显差异,血常规指标及生化指标也无明显差异,对两组小鼠的主要脏器进行病理组织学检查,两组小鼠的主要脏器经肉眼及镜检均无病理性差异。结论经实验测得小鼠灌胃给药的最大耐受量是300g/kg,相当于临床成人日剂量的181倍。益肾通络方浓缩合剂有较高的安全性。
Objective We study on the security of Yishen Tongluo concentrated decoction though observing mice's toxic reaction during 14d after given Yishen Tongluo decoction intragastieally. Methods The mice were given high dosage (2.5g/m1) and the most volume (0.4ml/10g), and were affused three times during 24h. Then mice's action and the number of deaths were recorded. There was no death. LD 50 could not be determined. We had no other choice to measure the maximal tolerance dose. Results Mice's in good conditions during 14d after administrating, the appetite and activity had no change, and there was no death. The weight, blood count and blood biochemical parameter had no distinct difference. The macroscopic observation and microscopic observation of two sets mice main vis- cera had no histopathological difference. Conclusion This experiment showed that maximal tolerance dose administration to mice was 300g/kg/d, about 181 times equal to the adult clinical daily dose. Yishen Tongluo concentrated decoction has higher security.
