摘要
目的:建立反相高效液相色谱法测定人体血浆中维拉佐酮的含量。方法:色谱柱为美国Diamonsil~(250 mm×4.6mm,5μm)C_(18)反相柱,流动相为乙腈-甲醇-缓冲液(30∶30∶40,V/V/V),流速为1.0mL·min^(-1),柱温30℃;检测波长240 nm。结果:维拉佐酮血浆浓度在0.16~32.00mg·L^(-1)范围内,线性关系良好(r=0.998 6,n=8),平均日间精密度为4.10%,平均相对回收率为97.85%。结论:本方法准确、快速、简便,适于人血浆中维拉佐酮的血药浓度检测和药动学研究。
OBJECTIVE To develop a reverse phase high performance liquid chromatography (RP-HPLC) for the determination of vilazodone in human plasma.METHODS A Diamonsil C18 column (250 mm×4.6 mm, 5 μm) was used with mobile phase composed of acetonitrile-methanol-buffer (30:30:40, V/V/V). Flow rate was 1.0 mL·min-1, column temperature was 30℃. UV detection wavelength was 240 nm.RESULTS In a mass concentration range of 0.16-32.00 mg·L-1, standard curve of vilazodone had a good linearity (r=0.998 6, n=8). The average daytime precision was 4.10%, the average relative recovery was 97.85%.CONCLUSION The established analytical method is accurate, rapid, convenient and suitable for the blood drug concentration detection and pharmacokinetics study of vilazodone in human plasma.
