摘要
目的探讨益心脑滴丸对肾性高血压大鼠(RHR)的降压作用及机制。方法构建两肾一夹(2K1C)经典肾性高血压大鼠模型,选取造模成功的大鼠并按收缩压水平分为模型组,益心脑高、中、低剂量组(2,1,0.5 g·kg^(-1)·d^(-1))及卡托普利组(25 mg·kg^(-1)·d^(-1)),同时设立假手术组,每组8只。灌胃给药每日1次,每周末测量血压,6周后处死并检测血浆内皮素(ET)和一氧化氮(NO),总抗氧化能力(T-AOC)、丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和肾组织活性氧(ROS)、谷胱甘肽过氧化物酶(GSH-Px)的变化;用酶联免疫法(ELISA)检测血浆肾素活性(RA)及血管紧张素Ⅰ(AngⅠ),AngⅡ的含量。结果益心脑滴丸能明显降低高血压大鼠收缩压(SBP)(P<0.05);增加血浆NO,降低ET含量(P<0.05);降低ROS和MDA的含量,提高抗氧化酶SOD,CAT和GSH-Px活性(P<0.05),使T-AOC增强;升高血浆AngⅠ含量,降低血浆AngⅡ含量(P<0.05)。结论益心脑滴丸有一定的降压作用,其机制可能与抗氧化,影响肾素-血管紧张素系统有关。
Objective To investigate the antihypertensive effect of Yixinnao dripping pills on renal hyperten-sive rats (RHR) and its mechanism. Methods The two-kidney one-clip (2K1C) goldblatt RHR models were estab-lished. The successfully established rat models were randomly divided into the model group, high, medium and low dose Yinxinnao groups (2, 1, 0.5 g·kg^-1·d^-1) and captopril group (25 mg·kg^-1·d^-1), and the sham operation group was established (n=8 each). All the rats received intragastric administration once daily and the blood pressure determina-tion every weekend, and were sacrificed at 6 weeks. The changes of plasma endothelin (ET), nitric oxide (NO), total anti-oxidation capability (T-AOC), malondialdehyde (MDA), superoxide dismutase (SOD), catalase (CAT), reactive oxy-gen species (ROS) of renal tissues, and glutathione peroxidase (GSH-Px) were determined. The levels of plasma renin activity (RA), angiotensin Ⅰ, Ⅱ (Ang Ⅰ, Ang Ⅱ) were measured by enzyme-linked immunosorbent assay (ELISA). Results Yixinnao dripping pills could significantly decrease the systolic blood pressure (SBP) of hypertensive rats (P〈0.05), increase the plasma NO and decrease the ET level (P〈0.05). T-AOC could be enhanced by decreasing the levels of ROS and MDA, and increasing the activities of antioxidant enzymes SOD, CAT and GSH-Px (P〈0.05). The level of plasma Ang I was increased, and the plasma Ang Ⅱ was decreased (P〈0.05). Conclusion Yinxinnao drip-ping pills may have a certain antihypertensive effect, and its mechanism may be related to antioxidant, affecting the renin-angiotensin system.
