摘要
为了研究伊犁马催乳素(PRL)基因、垂体转录因子(POU1F1)基因多态及其与产奶量之间的关系,试验以新疆伊犁昭苏牧场73头伊犁马母马为研究对象,利用PCR-RFLP技术分析了伊犁马PRL、POU1F1基因序列多态性,并与日均产奶量进行关联分析。结果显示,伊犁马PRL基因第二外显子上存在1个SNP:c.210C>T;POU1F1基因CDS区上没有发现多态,但内含子区存在1个SNP:g.2758G>A。POU1F1基因不同基因型间日均母马产奶量差异不显著(P>0.05),但PRL基因c.210位点多态对伊犁马日均产奶量有显著影响(P<0.05)。因此,PRL基因的多态位点有潜力用作伊犁马产奶量性状选育的分子标记。
In order to analyze the polymorphisms of prolactin (PRL) gene, pituitary transcription fac-tor 1 (POU1F1) and their association with milk yield of Yili horse, 73 Yili mares from Xinjiang Yili Zhao- su ranch were selected as samples to investigate the polymorphism of RRL, POU1F1 genes using PCR- RFLP technology, then performed the correlation analysis with daily milk yield. The results showed that no polymorphism was found in the CDS region of POU1F1 gene in Yili horse, but a SNP: c. 210 OT was found in the second exon of PRL gene; a SNP: g. 2758 G〉A was found in the intron of POU1F1 gene. Variance analysis results showed that no significant correlation (P〉0. 05) was found between the geno-types of POU1F1 gene (c. 143-262 G〉A) on daily milk yield in Yili horse, while the SNP in PRL gene showed the significant effect (P〈0. 05) on daily milk yield. Therefore, PRL gene is a potential useful marker in lactation performance selection of Yili horse.
