摘要
目的观察胰岛素致低血糖对糖尿病大鼠脑组织NGFmRNA表达变化的影响作用。方法48只Wistar大鼠按随机数字表法分为对照组(16只)、糖尿病模型组(32只),采用高脂高糖饮食+腹腔注射链脲佐菌素方法制备2型糖尿病大鼠模型,糖尿病模型组随机分为糖尿病非干预组(16只)和胰岛素低血糖组(16只)。胰岛素低血糖组大鼠予胰岛素皮下注射,每日1次,3组大鼠分别于第3、6、9、12天取4只大鼠的脑组织用RT-PCR法检测NGFmRNA。结果与对照组比较,糖尿病非干预组和胰岛素低血糖组大鼠脑组织NGF mRNA表达均不同程度下降,胰岛素低血糖组降低最为显著(P<0.01)。结论糖尿病降低了脑组织NGF mRNA表达,而胰岛素致低血糖能使脑组织NGFmRNA表达进一步下降,不利于糖尿病神经病变的再生修复。
Objective To observe the effect of hypoglycemia caused by insulin on NGF mRNA expression in rats brain with diabetes. Methods 48 adult Wistar rats according the random number table method were divided into control group (16 rats) and diabetes model group (32 rats). High fat and sugar dietase combinated + Streptozotocin intraperitoneal injection were used for making type 2 diabetes model. The diabetes model group was randomly divided into dia- bete non- intervention group (16 rats) and hypoglycemia caused by insulin group (16 rats). The rats in hypoglycemia caused by insulin group were injected insulin subcutaneously, once per day,. the brain tissues of 4 rats in 3 groups were detected NGF mRNA by the RT-PCR at 3rd, 6th, 9th and 12th day. Results Compared with control group, the expression of NGF mRNA in the brain tissues of diabete non- intervention group and hypoglycemia caused by insulin group were decreased at different degrees (P 〈 0.01), the hypoglycemia caused by insulin group was the most obvious (P 〈 0.01). Conclution Diabetes can effectively reduce the expression of NGF mRNA in brain tissue, hypoglycemia caused by insulin can make the expression of NGF mRNA further decreased, it is not conducive repairing diabetic neuropathy.
