健脾摄血方对ITP模型小鼠生命体征和脏器指数的影响 被引量:11

摘要 目的:观察健脾摄血方对ITP小鼠生命体征和脏器指数的影响,为临床应用提供支撑。方法:以抗小鼠血小板血清(APS)免疫造模法,建立BALB/c小鼠模型,体内造成ITP小鼠模型,观察强的松、健脾摄血方对ITP小鼠生命体征和胸腺、脾脏指数影响。结果:(1)各实验组小鼠体质量、体温与空白对照组比较,无统计学意义(P>0.05)。(2)造模第8天,模型组、强的松组、健脾组小鼠游泳时间与空白对照组比较,有统计学意义(P<0.01);实验结束后,模型组小鼠游泳时间仍低于空白组(P<0.05),强的松组、健脾组小鼠游泳时间均有所延长,以健脾摄血效果最佳。(3)从注射APS第4天开始,模型组、强的松组、健脾摄血组小鼠进食量明显减少,与空白对照组比较,具有统计学意义(P<0.05);干预给药第10天、第12天,强的松组模型小鼠进食量增加,与对照组比较,具有统计学意义(P<0.05);第16天健脾摄血组小鼠进食量增加,与模型组比较,具有统计学意义(P<0.05)。(4)从注射APS第6天开始,模型组饮水量增加,与空白组比较,具有统计学意义(P<0.05);给药第10天开始,强的松组小鼠饮水量增加,与空白组比较,具有统计学意义(P<0.05),而健脾摄血组小鼠饮水量无明显增加。(5)注射APS后由于免疫效应,模型组脾脏、胸腺明显增大,与空白组比较,具有统计学意义(P<0.05);经强的松干预后,脾脏、胸腺缩小,与模型组比较,具有统计学意义(P<0.01)。经健脾摄血方干预后,脾脏与模型组比较,无统计学意义(P>0.05);胸腺具有统计学意义(P<0.01)。结论:(1)注射APS复制的ITP小鼠模型符合病证结合模型特征。(2)强的松、健脾摄血方对ITP模型小鼠不耐劳力、食欲不振等脾气虚症状有明显改善效果。 Objective : To investigate the effect of Jianpi Shexue prescription on the vital signs and organ index of ITP mice models, providing support for clinical application. Methods : The ITP models of BALB/c mice were established by injec- tion of the anti - platelet serum (APS). Prednisone and Jianpi Shexue prescription were given, and then the changes of vital signs, the spleen and thymus indexes were detected. Results: (1)Mice body weight, body temperature had no sig- nificant change between the experimental group and control group ( P 〉 0. 05 ). (2)On the 8th day, the swimming time between control group and ITP, prednisone, Jianpi Shexue prescription group had statistically significant differences ( P 〈 0.01 ) ; after the experiment, the swimming time of ITP group was still lower than that of the control group ( P 〈 0. 05 ) , and was prolonged in prednisone group and Jianpi Shexue prescription group, Jianpi Shexue prescription group had the best effect. (3)From the 4th day, compared with the control group, the food - intake of ITP group, prednisone group and Jianpi Shexue prescription group was decreased obviously (P 〈 0.05 ) ; on the 10th and 12th days, the food - intake of prednisone group was increased significantly compared with that of control group (P 〈 0. 05 ) ; on the 16th day, the food - intake of Jianpi Shexue prescription group was increased obviously compared with that of ITP group ( P 〈 0. 05 ) ; (4)From the 6th day, the water consumption of ITP group was increased markedly compared with that of control group ( P 〈 0.05 ) ; From the 10th day, the water consumption of prednisone group was increased significantly compared with that of control group ( P 〈 0.05 ) ; the water consumption of Jianpi Shexue prescription group had no significant difference. (5)After administration of APS, the volume of spleen and thymus was enlarged obviously compared with that of control group (P 〈 0. 05 ) ; after administration of prednisone, the volume of spleen and thymus was reduced markedly compared with that of ITP group ( P 〈 0.01 ) ; the volume of spleen had no significant difference ( P 〉 0.05 ) and thymus was changed significantly in Jianpi Shexue prescription group (P 〈 0.01 ). Conclusion: (1)The ITP model was consistent with the characteristics of the syndrome model. (2)Prednisone and Jianpi Shexue prescription can effectively improve the symptoms of easy fatigue, loss of appetite and other symptoms of spleen Qi deficiency.
