益母草愈伤组织诱导与生物转化能力初探 被引量:2

摘要 目的:建立益母草愈伤组织培养体系,用于生物转化外源底物初探。方法:以MS为基本培养基,研究不同外植体及植物生长调节剂对益母草愈伤组织诱导及增殖的影响,并对获得的愈伤组织生物转化能力进行初探。结果:益母草子叶诱导率85.79%、真叶诱导率97.88%、下胚轴诱导率29.78%;诱导的最适配方为MS+2,4-D 0.5 mg/L+6-BA 2 mg/L;初期增殖最适配方为MS+2,4-D 0.5 mg/L+6-BA 2 mg/L;长期继代最适配方为MS+2,4-D 0.1 mg/L+6-BA 1 mg/L;最佳继代周期10 d;获得的3种愈伤组织悬浮培养均具有转化外源氢醌生成熊果苷的能力。结论:益母草愈伤组织经优选增殖,有望成为转化外源物质的良好材料。 Objective: To establish the callus culture system of Leonurus japonicus, which was used in bioconversion ex- ogenous substrates. Methods: MS was selected as the basic medium, and the effects were detected according to callus induction and proliferation with different explants and plant growth regulators, and then the callus ability of bioconversion was explored. Results: The callus induction rate of cotyledon was 85.79% ; the induction rate of true leaf was 97.88% ;the induction rate of hypocotyl was 29.78%. The optimum medium formula for inducing callus was MS + 2,4 - D 0.5 mg/L +6 - BA 2 mg/L; that for initial proliferation was MS +2,4 - D 0.5 mg/L +6 - BA 2 mg/L; that for callus Long -term successive transfer culture was MS + 2,4 -D 0.1 mg/L + 6 - BA 1 mg/L; the optimal cycle of subculture was 10 d; the three kinds of callus in suspension culture had the ability of hydroquinone into arbutin. Conclusion: By optimizing proliferation , the callus of Leonurus japonicus is expected to become a good material for transformation exoge- nous compounds.
