eNOS基因多态性与辽西地区汉族人群原发性高血压的相关性研究 被引量:1

摘要 目的拟通过对辽西地区汉族人群原发性高血压患者e NOS基因的检测,探讨eNOS基因a/b多态与原发性高血压(Essential Hypertension,EH)之间的相关性。方法本研究将EH患者(217例)与血压正常人(206例)进行病例-同胞对照设计,应用聚合酶链反应(Polymerase Chain Reaction,PCR)与凝胶电泳实验方法,对其e NOS基因a/b多态进行检测分析和基因分型,数据分析采用SPSS 11.5统计软件。结果对所有研究对象的检测结果显示:EH组和对照组之间比较,e NOS基因a/b多态基因型与等位基因的频率分布具有明显差异(基因:χ2=3.812,P=0.016;等位基因:χ2=4.302,P=0.001)。对所有研究对象的检测数据进行单因素Logistic回归分析,显示as+ab基因型与EH具有显著相关性,是EH发生的危险因素(OR=1.349,95%CI:1.032~2.006)。通过多因素Logistic回归分析,得出年龄因素、患者空腹血糖以及e NOS基因a/b多态三者是EH发生的独立危险因素。通过Meta分析显示辽西地区汉族人病例组与对照组的e NOS基因a/b多态aa+ab基因型频率分布具有明显差异,具有aa+ab基因型的人更易发生EH(OR=1.87,P<0.001)。结论 e NOS基因a/b的多态性与辽西地区汉族人群发生EH的风险密切相关。 Objective This paper is intended to find out the relationship between essential hypertension (EH) and eNOS a/b polymorphisms by testing eNOS genes in patients with essential hypertension in Han population in western Liaoning regions.Methods A case-sibling control design was conducted between EH patients (217 cases) and those with normal blood pressure (206 cases),and Polymerase Chain Reaction (PCR) and gel electrophoresis were applied to test and analyze eNOS a/b polymorphisms.SPSS 11.5 was used in this research for data analysis.Result The test results from all studied targets showed that eNOS a/b polymorphisms genotype was significantly different from the genotype frequency of alleles when comparing EH group with the control group (genes:X2=3.812,P=0.016;alleles:X2=4.302,P=0.01).Single-factor logistic regression analyses for the testing data of all targets showed that as+ab genotype had significant correlation with EH,which was a dangerous factor for EH (OR=1.349,95%CI:1.032~2.006).Multivariate logistic regression analysis demonstrated that ages,fasting blood-glucose and eNOS a/b polymorphisms were independent factors for EH.And Meta analysis showed that eNOS a/b polymorphisms genotype frequency distributions were different when comparing the patients group with the control group,and people with aa+ab genotype were more prone to EH (OR=1.87,P〈0.001).Conclusion eNOS a/b polymorphisms are closely related with the risk of developing EH among Han population in western Liaoning regions.
