芽孢皮层肽聚糖水解物RP-HPLC法分析条件优化

摘要 通过试验确定RP-HPLC法分析芽孢皮层肽聚糖水解物的最佳条件如下:在Welch Ultimate AQ-C18色谱柱上,柱温为30℃,流速为1 m L/min,检测波长为206 nm,洗脱时间为120 min,以甲醇和0.05 mol/L磷酸盐缓冲液(p H=4.31)做流动相,在0 min^5 min时使用100%磷酸盐缓冲溶液,在5 min^120 min时流动相中的甲醇浓度从0梯度增加到30%。结果表明:用RP-HPLC法分析芽孢皮层肽聚糖水解物是可行的,为研究高压热处理下芽孢皮层肽聚糖支架水解机理,并进一步阐明高压热处理杀灭细菌芽孢的机理提供了研究方法支撑。 By experiment, the optimal RP-HPLC conditions for analyzing spore's cortical peptidoglycan hy-drolysate were as follows:on Welch Ultimate AQ-C18 chromatographic column, column temperature 30℃, flow speed 1 mL/min, detection wavelength 206 nm, elution time 120 min, with methanol and 0.05 mol/L PBS (pH=4.31) as mobile phase, in 0 min-5 min 100%PBS was used, in 5 min-120 min the methanol concentra-tion increased gradually from 0 to 30%. The result indicated that it was feasible to use RP-HPLC for analyzing spore's cortical peptidoglycan hydrolysate, which provided research method support for studying the hydrolysis principle of spore's cortical peptidoglycan under HPTS (High Pressure Thermal Sterilization) treatment, and further for clarifying the principle of spore inactivation by HPTS.
