摘要
目的研究锌指蛋白703(ZNF703)在甲状腺乳头状癌中的表达(包括阳性率及相对表达量)及阳性率与临床特征的关系。方法应用免疫组织化学法(pv9000法)检测组织芯片(包括36例甲状腺乳头状癌及14例癌旁正常组织)中ZNF703的阳性表达率,并分析癌组织中的阳性表达率与芯片包含的相关临床资料(年龄、性别、肿瘤大小、淋巴结转移、临床分期)之间的关系。另收集20例临床新鲜甲状腺乳头状癌及匹配癌旁组织样本,应用Western blot及RT-PCR技术检测ZNF703在20对组织样本的蛋白和m RNA相对表达量。结果免疫组化提示ZNF703在甲状腺乳头状癌的阳性表达率为66.7%,较癌旁组织的21.4%明显增高,差异具有统计学意义(P<0.05);癌组织阳性表达率与年龄、性别无关,与肿瘤大小、临床分期、淋巴结转移有关(P<0.05)。Western blot及RT-PCR提示ZNF703在20例甲状腺乳头状癌组织的蛋白相对表达量[(0.893±0.113)vs(0.510±0.072)]及m RNA相对表达量[(6.399±2.096)vs(1.285±0.354)]均高于匹配癌旁组织,差异均有统计学意义(P<0.05)。结论 ZNF703在甲状腺乳头状癌中的表达高于癌旁组织,并且在癌组织中ZNF703阳性率高低与患者临床分期、淋巴结转移及肿瘤大小有关,可见ZNF703在甲状腺乳头状癌的发生及进展中可能发挥促进作用。
Objective To study the expressions(including the positive rate and the relative expression) of zinc finger protein 703(ZNF703) in papillary thyroid carcinoma and the relationship between the positive rate and the related clinical characteristics. Methods Immunohistochemistry PV-9000 method was used to detect the positive expression rate of ZNF703 in tissue chips(including 36 cases of papillary thyroid carcinomas, 14 cases of para-carcinoma tissues),and the relationship between the positive expression rates of ZNF703 in papillary thyroid carcinomas and clinical characteristics included by the chip(including age, gender, tumor size, lymph node metastasis, clinical stage) was analyzed. In addition, 20 cases of clinical fresh papillary thyroid carcinomas and para-carcinoma tissues were collected. Western blot and RT-PCR were used to detect the relative expressions of protein and m RNA of ZNF703 in 20 pairs of cases papillary thyroid carcinomas and para-carcinoma tissues. Results Immunohistochemistry study showed that the positive expression rate of ZNF703 in papillary thyroid carcinoma(66.7%) was significantly higher than that in para-carcinoma tissues(21.4%)(P〈0.05). The positive rate of papillary thyroid carcinomas was correlated with tumor size, lymph node metastasis, clinical stage(P〈0.05), and was not related to age, gender. Western-blot and RT-PCR indicated that the relative expression of protein and m RNA of ZNF703 in 20 cases of papillary thyroid carcinomas were respectively(0.893±0.113)and(6.399±2.096), which were significantly higher than(0.510±0.072) and(1.285±0.354) in matched para-carcinoma tissues(P〈0.05). Conclusion The expression of ZNF703 in papillary thyroid carcinoma is higher than that in para-carcinoma tissue, and the positive rate of ZNF703 in cancer tissues is correlated with clinical stage, lymph node metastasis and tumor size. So ZNF703 may play a carcinogenic role in the occurrence and development of papillary thyroid cancer.
