复合平肥茶对单纯性肥胖大鼠脂肪组织UCP-1 mRNA和下丘脑POMC mRNA的影响 被引量:1

Effect of Compound Pingfei Tea(CPFT,复合平肥茶) on the Expression of UCP-1 in Adipose Tissue and POMC in Hypothalamus of DIO Rats

摘要 目的:观察复合平肥茶对单纯性肥胖大鼠(DIO)体质量、血脂、血糖、白色脂肪组织解偶连蛋白1(UCP-1)和下丘脑阿片促黑色素原(POMC)的影响。方法:50只SD雄性大鼠随机分为低脂组(10只)和高脂组(40只),分别以低脂饲料及高脂饲料喂养,高脂组大鼠造模成功后,随机分为模型组、奥利司他组、复合平肥茶组,每组10只。低脂组和模型组予以蒸馏水灌胃,奥利司他组和复合平肥茶组分别予以奥利司他和复合平肥茶灌胃。治疗结束后,分别检测体质量、血脂、血糖、白色脂肪组织UCP-1 m RANA表达水平和下丘脑POMC mRNA表达水平。结果:复合平肥茶组和奥利司他组体质量增长明显低于模型组(P<0.05),甘油三酯(TG)、血总胆固醇(TC)、血低密度脂蛋白(LDL)、高密度脂蛋白(HDL),空腹血糖(FPG)均明显低于模型组(P<0.05);复合平肥茶和奥利司他组脂肪组织UCP-1 mRNA和下丘脑POMC mRNA较模型组均明显升高(P<0.05)。奥利司他组大鼠皮毛发黄,精神萎靡,活动度减少,且大便溏稀不成形,而复合平肥茶大鼠皮毛有光泽,精神可,活动灵敏,大便正常。结论:复合平肥茶可以抑制大鼠体质量的增长,增加UCP-1 mRNA和POMC mRNA表达促使白色脂肪棕色化,保护糖脂代谢平衡。 Objective: To observe the effects of the Compound Pingfei Tea on body weight, blood fat, fasting plasma glucose and UCP-1 mRNA and POMC mRNA expression in DIO rats. Methods: The 50 rats were ran- domly divided into low fat diet group (n=lO) and high fat diet group (n=40). After the DIO model was estab- lished, the successful model rats were randomly divided into model group, Orlistat group (OLST) and the Com- pound Pingfei Tea group (CPFT). Low fat diet group (LFD) and model group were treated with distilled water by gavage. OLST and CPFF were treated with orlistat and the Compound Pingfei Tea by gavage respectively. After 4 weeks, all rats were sacrificed to detect the body weight, blood fat, PFG, UCP-1 mRNA and POMC mRNA expression in adipose tissue and the adipocyte. Results: The OLST and CPFT group showed lower weight gain than model group (P〈0.05), as well as lower TG, TC, LDL, HDL and FPG than model group (P〈0.05). The OLST and CPFT group showed higher expression of UCP-1 mRNA and POMC mRNA than model group (P〈 0.05). The rats in OLST showed watery stools, a decrease in activity, mental fatigue and yellow hairs. But rats in CPFT showed regular stools and sleek coat. Conclusion: The Compound Pingfei Tea shows effects on weight loss increasing the UCP -1 mRNA and POMC mRNA expression, promoting the browning of white fat and pro- tecting the glucolipid metabolic balance.
