摘要
目的:从CD4^+ CD25^+ Foxp3^+ Treg细胞变化角度探讨安胎汤对先兆流产大鼠的影响及作用机制。方法:将大鼠雌雄以2∶1的比例配对后筛选出孕鼠48只,以缩宫素及针灸合谷、三阴交穴的方法建立先兆流产模型,随机分为6组,分别为正常妊娠组、安胎汤低剂量组、安胎汤中剂量组、安胎汤高剂量组、黄体酮组及先兆流产大鼠模型组。于妊娠第13天取大鼠外周血后即用流式细胞仪检测CD4^+ CD25^+ Foxp3^+ Treg细胞百分率,然后处死大鼠,计数双侧子宫着床点数及活胎数。结果:安胎汤可提高先兆流产孕鼠胚胎吸收率和CD4^+ CD25^+ Foxp3^+ Treg细胞百分率,降低胚胎吸收率(P<0.01)。结论:安胎汤通过调节先兆流产大鼠CD4^+ CD25^+ Foxp3^+ Treg细胞百分率达到保胎目的,且疗效与剂量相关。
Objective :To investigate the effect of Antai decoction on rats with threatened abortion and its mechanism of action from the perspective of the changes in CIM + CD25 + Foxp3 + Treg cells. Methods : Female and male rats were matched at a ratio of 2:1 ,and then 48 pregnant rats were selected. A rat model of threatened abortion was established by oxytocin and ac- upuncture at Hegu and Sanyinjiao points ,and the rats were randomly divided into normal pregnancy group ,low - ,middle - , and high -dose Antai decoction groups, progesterone group, and threatened abortion model group. Peripheral blood samples were collected on day 13 of pregnancy,and flow cytometry was used to measure the percentage of CIM + CD25 + Foxp3 + Treg cells. The numbers of implantation sites and live fetuses at both sides of the uterus were counted. Results : Antai decoction in- creased the percentage of CIM ~ CD25 + Foxp3 + Treg cells in pregnant rats with threatened abortion and reduced embryo re- sorption rate ( P 〈 0. 01 ). Conclusion : Antai decoction can increase the percentage of CD4 + CD25 + Foxp3 + Treg cells in preg- nant rats with threatened abortion and thus helps to prevent miscarriages in a close -dependent manner.
