雷公藤多苷对大鼠卵巢功能耗损的影响与菟丝子总提取物干预作用的实验研究 被引量:12

Effect of tripterygium glycoside on ovarian function loss in rats and interventional effect of total extract of China dodder seed: an experimental study

摘要 目的:观察雷公藤多苷(Tripterygium Glycosides,TG)对大鼠卵巢功能耗损的影响,并探讨菟丝子提取物对其卵巢功能的保护和修复作用。方法:将SD大鼠50只随机分为A、B1、B2、C1、C2共5组,每组10只。A为空白对照组,每天0.1m L 0.9%氯化钠注射液灌胃;B1、B2为TG给药组,于每天8∶0、20∶00分别灌胃TG 50mg/kg/d,连续给药14d;C1、C2为菟丝子提取物给药组,每天灌胃菟丝子提取物895.69mg/kg,给药2次,上午下午各1次,连续3d。HE染色观察卵巢组织形态,放射免疫法检测血清雌二醇(E_2)、促卵泡刺激素(FSH)、促黄体生成激素(LH)水平。结果:与A组比较,B1、B2组动情周期延长为7~8d/次,卵巢萎缩,结构受损。E_2、FSH、LH水平均无明显改变;而C1、C2组动情周期延长为7~8d/次,偶为5d/次,卵巢结构病变减轻,E_2、FSH及LH水平均无明显改变。结论:TG可致大鼠卵巢功能损害,晚上给药较白天给药损害较轻,菟丝子提取物对其生殖系统损害有明显改善作用。 Objective :To investigate the effect of tripterygium glycosides (TG) on ovarian function loss in rats and the protec- tive and repair effects of China dodder seed extract on ovarian function. Methods : A total of 50 Sprague - Dawley rats were random- ly divided into A,B1 ,B2, C1, and C2 groups,with 10 rats in each group. Group A was blank control group treated with 0. lmL 0. 9% sodium chloride injection by gavage daily. The rats in groups B1 and B2 were treated with TG 50mg/kg/d by gavage at 8:00 and 20:00 ,respectively ,and those in groups C1 and C2 were treated with China dodder seed extract 895.69 mg/kg by ga- vage in the morning and in the afternoon, respectively. The course of treatment was 3 days for all groups. HE staining was used to observe ovarian histomorphology and radioimmunoassay was used to measure the serum levels of estradiol (E2 ) , follicle -stimula- ting hormone (FSH) , and luteinizing hormone (LH). Results:Compared with group A, groups B1 and B2 had an extended estrous cycle of 7 - 8 days/time, as well as ovarian atrophy and structural damage, while there were no significant changes in the levels of E2, FSH, and LH. Groups C1 and C2 had an extended estrous cycle of 7 ~ 8 days/time, occasionally 5 days/time, and alleviated o- varian lesions, and there were no significant changes in the levels of E2, FSH, and LH. Conclusion : TG can cause ovarian function loss in rats, and administration in the evening helps to further alleviate such damage. China dodder seed extract has a significant effect in improving damage of the reproductive system.
