黄芩苷对ox-LDL诱导的人脐静脉内皮细胞损伤的保护作用 被引量:10

摘要 目的观察黄芩苷对氧化型低密度脂蛋白(ox-LDL)诱导的血管内皮细胞的保护作用。方法将体外培养的人脐静脉内皮细胞(EAhy926)分成对照组、ox-LDL组、黄芩苷组和不同浓度的黄芩苷(25、50和100 mg/L)+ox-LDL组,培养24 h。采用CCK-8检测细胞活力,ELISA检测细胞培养上清液中TNF-α和IL-6水平,Western blot检测凋亡相关蛋白Bax和Bcl-2的表达。结果与ox-LDL组相比,50、100 mg/L黄芩苷+ox-LDL组细胞存活率明显升高(P<0.05),细胞培养上清中TNF-α和IL-6水平均明显下降(P<0.05),Bax/Bcl-2比值降低(P<0.05)。结论黄芩苷对预防内皮细胞损伤的保护功能可能是通过抑制炎症因子,减少细胞凋亡,增强细胞活性实现的。 Aim To observe the protective effect of Baicalin on vascular endothelial cells induced by oxidized low density lipoprotein (ox-LDL). Methods In vitro cultured human umbilical vein endothelial cells (EAhy926) were divided into control group, ox-LDL group, Baicalin group and different concentrations of Baicalin (25, 50 and 100 mg/L) +ox-LDL groups, cultured 24 h. Cell viability was detected by CCK-8, ELISA was used to detect the level of IL-6 and TNF-α in cell culture supernatant, Western blot was used to detect the expression of apoptosis related protein Bax and Bcl-2. Results Compared to the ox-LDL group, the cell survival rates of 50, 100 mg/L Baicalin+ox-LDL groups were significantly increased (P〈0.05), the protein levels of both TNF-α and IL-6 in cell culture supematant were decreased significantly (P〈0.05), and the ratio of Bax/Bcl-2 decreased (P〈 0.05). Conclusion Baicalin protects against endothelial cell injury, which may be achieved by inhibiting inflammatory factors, reducing apoptosis and enhancing cell activity.
