调脂颗粒对房颤大鼠血栓前标志物DD和vWF的影响研究 被引量:2

摘要 目的研究调脂颗粒对心房颤动(房颤)大鼠血栓前标志物D-二聚体(DD)和血管性假血友病因子(vWF)的影响。方法以氯化钙-乙酰胆碱混合溶液经尾静脉注射,建立房颤大鼠模型。从中筛选具有典型房颤心电图的大鼠30只,随机分为房颤模型组、调脂颗粒治疗组、华法林治疗对照组,各10只,另平行设定健康对照组10只,连续给予相应药物治疗4周后,测定各组大鼠血清中DD和vWF水平。结果实验动物造模较成功。各药物治疗组都能对房颤大鼠的日常状态有一定改善。在房颤模型组大鼠注射造模药剂第7天后,其房颤持续时间与健康对照组比较,差异有统计学意义(P<0.05)。房颤模型组、调脂颗粒治疗组、华法林治疗对照组中经造模后大鼠呼吸频率显著上升,均较健康对照组高,差异有统计学意义(P<0.05)。经给药后,调脂颗粒治疗组、华法林对照组大鼠呼吸频率较房颤模型组降低,差异均有统计学意义(P<0.05)。房颤模型组大鼠血清中DD和vWF水平较健康对照组明显增加,差异均有统计学意义(P<0.05);调脂颗粒治疗组和华法林治疗对照组大鼠血清中DD和vWF水平明显低于房颤模型组,差异均有统计学意义(P<0.05);而调脂颗粒治疗组对房颤大鼠血清中DD和vWF的改善程度接近于法华林治疗对照组,差异均无统计学意义(P>0.05)。结论调脂颗粒可明显降低房颤大鼠血液中DD和vWF水平,可有效防治与治疗房颤,效果同华法林。 Objective To explore the influence of Tiaozhi granules on prethrombotic state molecular marker D-Dimer(DD) and von willebrand factor(vWF) of rats with atrial fibrillation(AF).Methods The AF rat model was established by intravenously injected with calcium chloride-acetylcholine mixture through caudal vein.Thirty rats with typical electrocardiogram of AF were selected and randomized into AF model group,Tiaozhi granules group and warfarin group,10 cases in each group;while 10 rats were selected as healthy control group.The levels of DD and vWF of rats in different groups were measured after 4-week successivly respective administration.Results The rat model was established successfully,and daily status of the AF rats in treatment groups all had some improvement.On the 7th day of medication,compared with the control healthy group,the time of AF duration in the rats model groups had statistically significant difference(P〈0.05).The respiratory rate of rats in AF model group,Tiaozhi granules group and warfarin group increased prominently after model establishment,comparing with the healthy control group,the difference had statistical significance(P〈0.05).After medication,the respiratory rates of the rats in Tiaozhi granules group and warfarin group were inferior to the AF model group with statistically significant difference(P〈0.05).The levels of serum DD and vWF in the AF model group were more than those in the healthy control group with statistically signifi-cant difference(P〈0.05);the levels of serum DD and vWF in Tiaozhi granules group and warfarin group were obviously lower than those in the AF model group with statistically significant difference(P〈0.05);while the improvement of serum DD and vWF in the Tiaozhi granules group were close to those in warfirin group without statistically significant difference(P〉0.05).Conclusion Tiaozhi granules can reduce the level of serum DD and vWF of the AF rats obviously,and it can prevent and treat AF effectively with equal efficacy to warfarin.
