摘要
[目的]通过观察乌头汤对糖尿病周围神经病变(DPN)大鼠胫神经神经生长因子(NGF)受体、酪氨酸蛋白激酶A(TrkA)和p75神经营养因子受体(p75NTR)表达及血清NGF含量的影响,探索该方改善DPN的可能机制。[方法]Wistar大鼠腹腔注射链尿佐菌素(STZ)建立糖尿病模型,观察乌头汤对胫神经纤维组织TrkA、p75NTR及血清NGF的影响。[结果]干预前后模型组和乌头汤组均较空白组FBG显著升高(P<0.01),各组内FBG在干预前后无统计学差异(P>0.05);与模型组比较,乌头汤组血清NGF含量明显升高(P<0.05),神经组织p75NTR明显降低(P<0.05)。乌头汤组p75NTR免疫组化染色阳性表达程度较模型组明显降低。[结论]乌头汤改善DPN的作用可能与其能够调节神经纤维p75NTR含量和血清NGF含量有关。
[Objective] To study the effect and mechanism of Aconite Main Tuber Decoction on NGF,TrkA and p75 NTR in rats developed diabetic peripheral neuropathy(DPN). [Methods] To set up pathological model of DPN, wistar rats were given streptozotocin(STZ) by intraperitioneal injection. NGF in serum and TrkA, p75 NTR in tibial nerve tissue were observed. [Results] Compared with blank control group, FBG were higher in both before and after the intervention groups and Aconite Main Tuber Decoction group( P〈0.01). No significant difference in FBG within the groups before and after intervention(P〉0.05). Compared with model control group, NGF in serum were higher and p75 NTR in tissue were lower in medicinal group(P〈0.05). In medicinal group,immunohistochemistry showed a lower degree positive staining compared with model control. [Conclusion] Aconite Main Tuber Decoction could alleviate DPN by regulating p75NTR/TrkA balance and rising NGF concentration.
