电针对短暂性脑缺血发作大鼠神经元形态学及细胞凋亡的影响 被引量:13

摘要 目的:探讨电针对短暂性脑缺血发作大鼠神经元形态学及细胞凋亡的影响。方法:Wistar雄性大鼠32只随机分为正常对照组、假手术组、模型组、针刺组,每组8只。采用四血管阻断法建立短暂性脑缺血发作大鼠模型。正常对照组、假手术组、模型组不给予任何治疗。针刺组造模后给予电针治疗,术后1周取材。尼氏染色观察神经元损伤情况,透射电镜观察神经元超微结构;原位末端标记法检测(TUNEL染色)大脑皮质、海马CA1区神经元凋亡。结果:神经元尼氏染色结果:模型组染色较浅,神经元尼氏染色阳性细胞数较少。而电针组较模型组神经元尼氏染色阳性细胞数增多(P<0.05)。电镜观察结果:模型组大脑皮层、海马CA1区见神经细胞肿胀,核周间隙扩张或消失,染色质凝集成块或溶解,线粒体嵴断裂;电针组电镜下观察线粒体肿胀轻微,毛细血管水肿少见,胞浆内细胞器丰富,结构完整。细胞凋亡检测结果:正常对照组大脑皮层、海马CA1区假手术组凋亡阳性细胞较少见,模型组大脑皮层、海马CA1区可见较多凋亡细胞,针刺组与模型组比较,凋亡细胞明显减少,具有显著性差异(P<0.05)。结论:电针可以减轻短暂性脑缺血发作大鼠大脑皮层及海马CA1区神经元损伤,抑制神经元凋亡。 Objective: To investigate the effect of electroacupuncture on the morphology and apoptosis of neurons in rats with transient ischemic attack. Methods: Thirty-two male Wistar rats were randomly divided into a normal control group,a sham operation group,a model group and an acupuncture group,with 8 rats in each group. A rat model of transient ischemic attack was established by four vessel occlusion. Normal control group,sham operation group and model group were not given any treatment,and electroacupuncture group was given electroacupuncture treatment. Nissl bodies were observed by Nissl's staining. The ultrastructure of neurons was observed by the electron microscope,and neuronal apoptosis was detected by in situ cell apoptosis( TUNEL staining) in the cerebral cortex and hippocampus CA1 area. Results: The number of positive cells in the electroacupuncture group was more than that in the model group( P〈0. 05). The result of electron microscope showed that swelling of the nerve cells,the expansion or disappearance of the nuclear gap,chromatin condensation into blocks or dissolution and mitochondrial crest fracture were found in the cerebral cortex,hippocampal CA1 area in the model group. But slight swelling of mitochondria and capillary edema,abundant and complete structure of cytoplasmic organelles were found in electroacupuncture group. TUNEL staining showed that apoptosis cells were rare in the cerebral cortex and hippocampus CA1 area in the normal control group and sham operation group. Compared with the model group,the apoptosis cells in the electroacupuncture group were reduced,and there was a significant difference between two groups( P〈0. 05). Conclusion: Electroacupuncture can allieviate neuronal damage and inhibit neuronal apoptosis in cerebral cortex and hippocampus CA1 area in rats with transient ischemic attack.
