慢性疼痛对孕鼠及其子代抑郁行为的影响

Effects of chronic pain on the depression behavior of pregnant mice and their offspring

目的:构建孕鼠慢性疼痛动物模型,观测慢性疼痛对孕鼠及其子代抑郁行为的影响。方法:将交配成功的母鼠随机分为正常对照组、假手术对照组和慢性疼痛组(n=10)。慢性疼痛组母鼠足跖关节皮内注射无菌完全弗氏佐剂100μl[500μg·(100μl)-1]诱导产生关节炎痛,疼痛可覆盖母鼠整个孕期(22 d左右)及产后1周。用糖水消耗实验、旷场实验在不同时间点分别检测母鼠及子鼠的抑郁行为表现。结果:(1)母鼠:在造模后第12、18天及产后第6天,糖水实验中慢性疼痛组小鼠糖水偏好度与假手术对照组比较均显著降低(P<0.01),旷场实验中慢性疼痛组小鼠直立次数和中央区活动距离与假手术对照组比较均显著降低(P<0.05或P<0.01),余时间点3组各指标相比差异无统计学意义(P>0.05)。(2)子鼠:慢性疼痛组子鼠在出生后3周时其糖水偏好度、直立次数、中央区活动距离均低于假手术对照组(P<0.05或P<0.01),在出生后6周时其糖水偏好度及直立次数低于假手术对照组(P<0.05或P<0.01),余时间点3组各指标相比差异无统计学意义(P>0.05)。结论:慢性疼痛对孕鼠及其子代的抑郁行为有促发作用。

Objective： To construct the animal model of chronic pain in pregnant mice,observe the effects of chronic pain on the depression behavior of pregnant mice and their offspring. Methods： The pregnant mice were divided into control group,sham-operation control group and chronic pain group（ n = 10）. In the chronic pain group,aseptic complete Freund’s adjuvant 100 μl [500 μg·（ 100 μl）-1]was injected intracutaneously in the paw joint of pregnant mice to induce arthritis pain. The pain could cover the whole pregnancy（ about 22 days） and 1 week after delivery. With syrup consuming experiment and open field test,the depression behavior of pregnant mice and their offspring was tested at different time points. Results： On the 12 th day,18 th day after making model and the6 th day after delivery,in the syrup consuming experiment,the sucrose preference degree of the female mice in the chronic pain group was significantly lower than that of the sham-operation control group（ P 〈 0. 01）,and in the open field test,the straight times and the movement distance in the central quadrant of the female mice in the chronic pain group was significantly lower than that of the sham-operation control group（ P 〈 0. 05 or P 〈 0. 01）. The index of three groups had no difference at other time points（ P 〉 0. 05）. When the child mice were 3 weeks,their sucrose preference degree,straight times and movement distance in the central quadrant of the chronic pain group was lower than those of the sham-operation control group（ P 〈 0. 05 or P 〈 0. 01）. When the child mice were 6 weeks,their sucrose preference degree and straight times of the chronic pain group was lower than those of the sham-operation control group（ P 〈 0. 05 or P 〈 0. 01）. The child mice’ index of three groups had no difference at other time points（ P 〉0. 05）. Conclusion： Chronic pain could spur the depression behavior of pregnant mice and their offspring.
