乳腺癌患者外周血中TGF-β1和P53蛋白与临床特征的关系 被引量:7

摘要 目的研究乳腺癌患者在治疗前后外周血中转化生长因子β1(TGF-β1)与P53蛋白含量变化,初步探讨二者与乳腺癌患者各项临床特征之间的关系。方法选取乳腺癌患者80例作为乳腺癌组,30例乳腺增生女性作为乳腺增生组。通过酶联免疫吸附试验分别测定乳腺癌患者于治疗前后外周血中TGF-β1与P53蛋白含量变化,与乳腺增生组对照,说明其治疗前后的浓度变化及与乳腺癌各项临床特征之间的关系。结果乳腺癌患者中外周血中TGF-β1及P53蛋白的含量显著高于乳腺增生组(P<0.05),且经过有效的治疗后,二者的含量显著下降(P<0.05)。TGF-β1与P53蛋白的含量在肿瘤的组织学分级及腋下淋巴结转移与否有显著的关联(P<0.05),与乳腺癌组织中的雌激素受体表达无显著相关(P>0.05)。乳腺癌患者的周血中TGF-β1与P53蛋白含量呈正相关(r=0.348,P<0.01)。结论乳腺癌患者外周血中TGF-β1与P53蛋白的含量显著增高,且与肿瘤组织学分级及腋下淋巴结是否转移显著相关,能够为乳腺癌的临床治疗工作提供新的靶点及思路。 Objective To observe the changes of TGF-β1,P53 protein levels in peripheral blood of patients with breast cancer in order to explore the correlation between the two factors and clinical characteristics of nreast cancer. Methods Eighty patients with breast cancer were enrolled as breast cancer group,and 30 patients with hyperplasia of mammary glands were served as mammary glands hyperplasia group( MGH group). The expression levels of TGF-β 1 and P53 protein in peripheral blood of patients with breast cancer before and after treatment were detected by enzyme linked immunosorbent assay( ELISA),which were compared with those in patients with hyperplasia of mammary glands. Moreover the correlation between the changes of expression levels of TGF-β1,P53 protein before and after treatment and clinical characteristics of breast cancer was analyzed. Results The expression levels of TGF-β1 and P53 protein in breast cancer group were significantly higher than those in MGH group( P〈0. 05). After treatment,the expression levels of TGF-β1 and P53 protein were obviously decreased( P〈0. 05). The expression levels of TGF-β1 and P53 protein were closely correlated to histological grading and axillary lymph node metastasis of breast cancer( P〈0. 05),however,which were not related with the expression levels of estrogen receptor in breast cancer tissues( P〉0. 05). Moreover the expression of TGF-β1 was positively correlated to that of P53protein( r = 0. 348,P〈0. 01). Conclusion The expression levels of TGF-β1and P53 protein in peripheral blood of patients with breast cancer are obviously increased,moreover,which are closely correlated to histological grading and axillary lymph node metastasis of breast cancer,therefor,which can provide new targets and ideas for clinical treatment of breast cancer
