摘要
采用十二烷硫酸钠—聚丙烯酰胺凝胶电泳法 (SDS -PAGE)和酸性聚丙烯酰胺凝胶电泳法(APAGE) ,分别对节燕 97- 1和加拿大优质面包小麦Glenlea及其杂交后代 (F3)的 30个株系进行高分子量麦谷蛋白亚基 (HMW -GS)和醇溶蛋白的遗传多样性分析。结果表明 ,Glenlea具有 7+8、5 + 10亚基 ,节燕 97- 1具有 1、7+ 9、5 + 10亚基 ,其杂交后代在 1A、1B、1D 3个位点上都具有多态性 ,共出现 10种亚基类型 ,其中有 5 + 12、5 + 10 + 12、7、6 + 8、7+ 8+ 9、2 0几种新亚基类型。普通小麦中极其稀少的 5 + 12亚基在此后代中占 13.3% ,5 + 10 + 12亚基占 10 % ,具有优质亚基组合 7+ 8、5 + 10的占 5 0 %以上。两个亲本的醇溶蛋白谱带也表现出不同带型 ,其杂交后代出现17种带型 ,在α、β、γ 3个区均有较大的差异。说明杂交后代的贮藏蛋白变异丰富 。
SDS-PAGE and APAGE analysis were used in high molecular weight glutenin subunit and gliadin variation of 30(F 3) hybrid progeny and their parents (Jieyan97-1 and Canada good bread wheat Glenlea). The results showed that Glenlea containing 5+10 and 7+8 subunits, Jieyan 97-1 containing subunits 1,7+9,5+10. In hybrid generation, the loci Glu-A1, Glu-B1 and Glu-D1 all showed variation. There are 10 different glutenin subunits in the 30 F 3 hybrid lines, of which 6 kinds new subunits of 5+12,5+10+12,7,6+8,7+8+9,20 were found, about 13.3% lines containing 5+12 subunit,10% lines containing 5+10+12 subunit, over 50% lines containing the high quality subunit combination of 7+8 and 5+10. There are different gliadin patterns in two parents, and 17 different patterns in hybrid generation. In α-zone,β-zone and γ-zone, the alleles of gliadin also showed variation. These results showed high genetic diversity of wheat storage proteins.
