摘要
目的:研究从明党参根皮超临界提取物中分得的5个香豆素化合物对体外培养的人脐静脉内皮细胞(HUVEC)氧化应激损伤的保护作用,以为其在动脉粥样硬化疾病上的运用提供理论依据。方法:以过氧化氢(H_2O_2)为氧化剂建立HUVEC细胞氧化应激损伤(OSI)模型,细胞种板经过24h孵育贴壁后,将细胞分为正常对照组、H_2O_2氧化损伤组、实验药物组,其中实验药物组给予药物预培养24h后,加入500μmol·L^(-1) H_2O_2,继续培养4h后MTT法检测细胞存活率。结果:5种呋喃香豆素均能改善H_2O_2所致的HUVEC细胞形态学损伤,提高细胞存活率,其中欧前胡素效果最好,能使细胞存活率提高22%。结论:5种呋喃香豆素具有保护血管内皮细胞免受过氧化损伤的作用。
Objective: To study the protective effects of five coumarins compounds from the supercritical fluid extract of Changium smyrnioides Wolff root bark on oxidative stress injury of cultured human umbilical vein endothelial cells (HUVECs) in vitro, to provide a theoretical basis for its application in atherosclerotic disease. Methods: The oxidative stress injury (OSI) model of HUVEC cells was established by using hydrogen peroxide (H2O2) as oxidant. After 24 hours incubation, the cells were divided into normal control group, H2O2 oxidative injury group, experimental drug group. After 24 h of incubation, 500μmol·L-1 H2O2 was added to the experimental group. The cell viability was measured by MTT assay after 4 h. Results: All the five furocoumarins could improve the morphological damage of HUVEC cells induced by H2O2, and the cell viability was improved. The effect of imperatorin was the best, and the cell survival rate was increased by 22%. Conclusion:The five furocoumarins have protective effects on endothelial cells from peroxidative injury.
