紫花牡荆素对佐剂性关节炎小鼠足肿胀度和细胞因子的影响

Effects of casticin on swelling of voix pedis and cytokines in adjuvant arthritis mice

目的研究紫花牡荆素对佐剂性关节炎小鼠血浆炎症相关细胞因子的作用及其与足跖肿胀度的相关性。方法 36只Bal b/c小鼠,随机分为6组:空白对照组、模型组、阳性药物组、紫花牡荆素12.5,25.0和50.0mg·kg^(-1)剂量组;小鼠右足垫皮内注射弗氏完全佐剂后连续灌胃给药12d,每日1次。第13天测量小鼠后足爪的肿胀度,第14天采血,用流式细胞仪的微球阵列(CBA)法检测佐剂性关节炎小鼠外周血中的致炎性细胞因子(IL-2、IFN-γ和TNF-α)和抗炎性细胞因子(IL-4和IL-5)的水平。结果紫花牡荆素可剂量依赖性地抑制Bal b/c小鼠后足爪的肿胀度;明显降低致炎性Th1细胞因子IL-2、IFN-γ和TNF-α的水平,显著提高抗炎性Th2细胞因子IL-4和IL-5的水平;小鼠后足爪肿胀度与血清细胞因子IL-2、IFN-γ和TNF-α的水平呈正相关,与IL-4和IL-5的水平呈负相关。结论紫花牡荆素改善佐剂性关节炎小鼠足跖肿胀度,可能是通过增加血液IL-4和IL-5含量,降低IL-2、IFN-γ和TNF-α含量,调节细胞因子平衡的结果。

Objective To study the effect of casticin on the cytokines and the swelling of voix pedis in adjuvant arthritis mice.Methods 36 Bal b/c mice were divided into 6groups randomly including the blank control group,the model group,the standard drug group,and the casticin group at the dose of 12.5,25.0and 50.0mg·kg-1,respectively.The mice after sub-plantar injection of Freund′s solution were performed to induce adjuvant arthritis.Drugs,once daily,were continuously administered for 12 days.On the13 th day after the adjuvant injection,the volume of the injected hind paw was measured,meanwhile the levels of pro-inflammatory cytokines IL-2,IFN-γand TNF-αand anti-inflammatory IL-4and IL-5in serum were detected using Cytometric Bead Array（CBA）in each group on the 14 th day.Results Casticin orally significantly suppressed the paw edema of Bal b/c mice,decreased the levels of pro-inflammatory Th1cytokines（IL-2,IFN-γand TNF-α）and increased the levels of anti-inflammatory Th2cytokines（IL-4and IL-5）in a dose-dependent manner.The swelling of voix pediswas positively correlated with the levels of IL-2,IFN-γand TNF-α,and was negatively correlated with the levels of IL-4and IL-5.Conclusion Casticin improves the swelling of voix pedis in adjuvant arthritis mice,the underlying mechanism of which is associated with the suppression of the levels of IL-2,IFN-γand TNF-α,and the enhancement of IL-4and IL-5contents,as well as keeping a balance of these cytokines.
