阻断磷脂酶C-γ1信号通路对卵巢癌细胞凋亡的影响

摘要 目的观察阻断磷脂酶C(PLC)-γ1信号通路对卵巢癌细胞凋亡的影响。方法用化学阻断剂U73122处理卵巢癌细胞,光镜观察细胞形态学改变,MTT法评价细胞生长抑制水平,流式细胞仪分析细胞周期变化及细胞凋亡率。结果磷脂酶C信号通路被阻断后,卵巢癌细胞出现明显的细胞凋亡形态学改变,细胞存活率显著下降,流式细胞仪检测到凋亡细胞,与浓度及时间呈一定关系。结论阻断磷脂酶C-γ1能启动卵巢癌细胞的凋亡。 Objective To explore the role of phospholipase C-γ1(PLC-γ1) signaling pathway in the apoptosis of SKOV3 cells.Methods Treatment of ovarian cancer cells with chemical blocker U73122,then observed the morphological changes of the cells under light microscope,MTT assay was used to evaluate the cell viability,and the percentage of apoptotic cells was analyzed by flow cytometry.Results After phospholipase C signaling pathway was blocked,ovarian cancer cells showed significant morphological changes in apoptosis,cell viability decreased significantly,flow cytometry was used to detect apoptotic cells,with the concentration and time was a certain relationship.Conclusion Blocking PLC-γ1 can initiate ovarian cancer cell apoptosis.
