齐墩果酸对体外培养人毛囊干细胞增殖的影响 被引量:1

摘要 目的:探究齐墩果酸对人毛囊干细胞增殖活性的影响。方法:取术中毁弃的人全层头皮标本,使用两步酶消化法分离提取毛囊干细胞进行体外培养,运用免疫荧光K15染色鉴定;将细胞以5×10~4个/mL的密度接种于96孔板,培养液中分别加入1×10^(-4)、1×10^(-3)、1×10^(-2)、0.1、1μg/mL梯度浓度齐墩果酸,于处理的24 h及48 h使用MTT法检测毛囊干细胞活性。结果:获得的毛囊干细胞呈典型的铺路石状生长,免疫荧光K15阳性,阳性率为(86.7±5.2)%(n=4)。与空白组相比,1×10^(-4)μg/mL及1×10^(-3)μg/mL齐墩果酸处理组在24 h及48 h的OD_(490)均有明显升高(P<0.05)。结论:齐墩果酸能够促进体外培养的人毛囊干细胞增殖。 Objective:To investigate the effect of oleanolic acid (OA) on proliferation of hu- man hair follicle stem cells (HFSC). Methods: HFSC were isolated and cultured in vitro and i- dentified by fluorescent staining. OA was added to medium at final concentration of 1 x 10-4 μg/ mL, 1 x 10-3 μg/mL, 1 x 10-2 μg/mL, 0. 1 μg/mL and 1 μg/mL, then MTT assay was per- formed when the cell culture was treated with OA at 24 h and 48 h to assess the number of viable cells. Results:Cultured cells were multipolar in shape and grew like typical cobble stone. The staining of K15 was positive, which is an epithelial stem cell marker. Positive rate of K15 is (86.7 -5.2)% (n =4). Compared to the vehicle control group, 0D490 of 1 X 10 -4 μg/mL and 1 x 10 -3 vg/mL of OA treated group were significantly increased ( P 〈 0. 05 ). Conclusion: OA could stimulate proliferation of human hair follicle stein cells in vitro.
