摘要
【目的】为研究郑州市区草坪白三叶草根瘤菌的遗传多样性。【方法】实验采用包括16S rDNA及IGS PCR-RFLP和16S rDNA基因序列分析方法。【结果】该研究从郑州市4个不同采样地点的草坪中采集白三叶草根瘤,并分离得到根瘤菌40株;经16S rDNA PCR-RFLP分析,初步确定所有供试菌株属于同一个属。而IGS PCR-RFLP分析将供试菌株分为四个类群,且来自郑州大学的供试菌株具有更加丰富的遗传多样性。对不同IGS型代表菌株的16S rDNA基因序列分析结果与16S rDNA PCR-RFLP的结果一致,表明所有供试菌都属于根瘤菌属(Rhizobium)。【结论】一共分离得到根瘤菌40株,它们都属于根瘤菌属,且分离自郑州大学的根瘤菌具有更加丰富的多样性。
[Objective]The objective of this study was to understand the genetic diversity of white clover rhizobial strains in Zhengzhou.[Methods]16S rDNA and IGS PCR-RFLP and 16 S rDNA gene sequence analysis were conducted in the research.[Results]40 strains were isolated from four different sampling sites in Zhengzhou.The phylogenetic analysis.The results of IGS PCR-RFLP showed that there were four PCR-RFLP fingerprint patternresults of 16 S rDNA PCR-RFLP showed that all of the isolates belonged to the same genus of Rhizobium ingroups and the isolates from the new campus of Zhengzhou University were with richer genetic diversity than other sites.According to the results of 16 S rDNA gene sequence analysis,all of the isolates were identified as Rhizobium strains which is consistent with the results of 16 S rDNA PCR-RFLP.[Conclusion] In this study,40 strains totally were isolated from Zhengzhou and they were identified as the genus of Rhizobium.And it is with much richer diversity of rhizobia from the new campus of Zhengzhou University.
