摘要 为了解决细叶石斛自然繁殖率低、市场种源短缺的问题,采用细叶石斛种子为材料,通过无菌萌发、原球茎增殖、原球茎分化和生根壮苗等离体快速繁殖过程获得组培苗.实验结果表明:1/2 MS+6%马铃薯粉培养基适合于原球茎形成和增殖;1/2 MS+6%马铃薯粉+0.8%花宝1号培养基可用于细叶石斛的原球茎分化和生根壮苗.共培养6~8月,细叶石斛组培苗的株高、茎粗、分蘖数、叶片数、根数和根长等指标分别达到4.43 cm、0.16 cm、1.76株、8.20片、5.48条和2.16 cm,而且栽后成活率可达97.94%. The natural reproduction rate of Dendrobiumhancockii Rolfe was low and the lack of provenance on the market. It used the seeds of D. hancockii Rolfe as material and methods of aseptic germination, protocorm multiplication and differentiation, rooting and growth-promoting to assist in solving these problems. The results show that the culture medium 1/2 MS+6% potato starch is suitable for formation and muhiplication of protocorm. 1/2 MS+6% of patatostarch and 8% of No.1 Huabao can be used for the differentiation of protocorm and rooting and growth-promoting. The plant height, stem diameter, tiller number, leaf number, root number, root length of tissue culture seeding of D. hancockff Rolfe are 4.43 cm, 0.16 cm, 1.76, 8.20, 5.48 and 2.16 cm respectively,and the survival rate is 97.94% after transplanting.
