摘要
研究微波引起白血病耐药细胞株K5 6 2 /MDR的凋亡以及对热休克蛋白 70表达的影响 .使用MTT方法检测细胞存活率 ,AnnexinⅤ /PI双参数法检测K5 6 2 /MDR细胞的凋亡率 ,流式细胞仪检测热休克蛋白 70的表达 .结果表明 ,微波可引起K5 6 2 /MDR细胞的凋亡 ,经 0 .2 0 ,0 .6 0W /cm2 功率密度微波作用 2 0min(保持 4 3℃ ) ,37℃培养 2 4h后 ,细胞凋亡率分别为 10 %、13.6 % ,而水浴加热引起的凋亡率仅为 3.8% .微波照射影响细胞内热休克蛋白 70的表达 ,在 4 3℃时 ,0 .2 0 ,0 .6 0W /cm2 功率密度微波作用 2 0min抑制HSP70的表达 .实验说明微波能引起K5 6 2 /MDR细胞的凋亡 。
The relationship between the induced apoptosis and expression of HSP70 in human multidrug-resistant leukemic cell line K562/MDR by microwave was investigated. Cell surviving fraction was determined by MTT assay. Apoptosis in K562/MDR was detected by Annexin Ⅴ/PI flow cytometrical dual parameter method. Expression of HSP70 was detected by flow cytometry. After 24 hours treatment by microwave(0.20,0.60 W/cm 2 for 20 min and keep in 43 ℃), the apoptotic rates were 10%(0.20 W/cm 2) and 13.6%(0.60 W/cm 2 ). The apoptotic rate of water bath at 43 ℃ for 20 min was only 3.8%. After 3 h treatment by microwave, the expression of HSP70 was lower than the control. Microwave could induce apoptosis in K562/MDR. It is believed that the apoptosis may be related to the decrease of HSP70.
