微囊藻毒素人工完全抗原制备与鉴定

Preparation and identification of artifical antigen of Microcystin-LR

摘要 为了研究微囊藻毒素-LR(MC-LR)的免疫学检测方法,试验制备了MC-LR人工完全抗原,采用水溶性碳二亚胺法将微囊藻毒素的羧基与牛血清白蛋白(BSA)的氨基偶联,同时利用琼脂糖凝胶电泳、非变性聚丙烯酰胺凝胶电泳、紫外扫描分析及ELISA等技术对偶联结果进行鉴定与分析。结果表明:微囊藻毒素的羧基与BSA的氨基偶联成功。说明人工完全抗原制备成功,可用于MCLR的免疫学检测。 In order to develop immunoassay methods for MC -LR, the MC -LR artificial complete antigen was prepared. The carboxy group of microcystin was conjugated with amino group of bovine serum albumin (BSA) by using carbodiimide method ; the prepared MC - LR antigen was verified and analyzed by using agarose gel electrophoresis, polyacrylamide gel electrophoresis, Uv scanning analysis, and EI.SIA technology. The results demonstrated that the carboxyl group of microcystin was successfully conjugated with bovine serum albumin (BSA). This data indicated that the artificial complete antigen was successfully prepared and had a potential application for the immunological detection of MC - LR.
