摘要
为了研究铁皮石斛对环磷酰胺(cyclophosphamide,CY)所致免疫抑制模型小鼠血清中抗氧化酶活性及细胞因子含量的影响,试验选用100只雌性KM种小鼠随机分为5组,每组5个重复,每个重复4只。Ⅰ组为对照组,每天灌服0.3 m L纯化水;Ⅱ组为模型组,每天灌服0.3 m L纯化水;Ⅲ、Ⅳ、Ⅴ组为试验组,分别灌服1,2,3 g/kg铁皮石斛水煎剂。Ⅱ、Ⅲ、Ⅳ、Ⅴ组按体重100 mg/kg腹腔注射CY,隔日注射1次,连续15 d。结果表明:试验成功构建免疫抑制小鼠模型。与Ⅰ组(对照组)相比,Ⅱ组(模型组)、Ⅲ、Ⅳ、Ⅴ组小鼠血清中GSH-Px活性显著降低(P<0.05);各组间CAT活性差异均不显著(P>0.05);Ⅱ组(模型组)和Ⅲ、Ⅳ、Ⅴ组小鼠血清中IL-4、IFN-γ、IL-10含量均存在显著差异(P<0.05)。与Ⅱ组(模型组)相比,Ⅰ组(对照组)与和Ⅲ、Ⅳ、Ⅴ组小鼠血清中MDA含量显著降低(P<0.05);Ⅰ组(对照组)和Ⅴ组小鼠血清中SOD活性显著升高(P<0.05),Ⅲ、Ⅳ组小鼠血清中SOD活性有升高趋势(P>0.05);Ⅲ、Ⅳ、Ⅴ组小鼠血清中IL-2含量显著降低(P<0.05);Ⅰ、Ⅲ、Ⅳ及Ⅴ组小鼠血清中IL-4/IFN-γ比值差异显著(P<0.05)。说明铁皮石斛能通过调控CY构建的免疫抑制小鼠血液内细胞因子,从而对免疫抑制小鼠的血清抗氧化酶活性及Th1与Th2细胞发挥调节作用,使得血液细胞因子指标恢复正常,有效提高环磷酰胺所致的小鼠免疫调节机能,具有一定的免疫增强作用。
In order to investigate the effect of Dendrobium officinale on activity of antioxidant enzymes and cytokines of the serum of CY caused by immunosuppressive mouse model,100 KM female mice were selected randomly divided into 5 groups in which each group has 5 repetitions,each repetition has 4. GroupⅠwas the control group,feeding with 0. 3 m L distilled water everyday; group Ⅱwas the immunosuppression model group,feeding with 0. 3 m L distilled water everyday; group Ⅲ,Ⅳ,Ⅴ were experimental groups,feeding with 1,2,3 g/kg different concentrations of Dendrobium officinale; group Ⅱ,Ⅲ,Ⅳ,Ⅴ were injected CY in accordance with 100 mg/kg body weight by intraperitoneal once every other day for 15 consecutive days. Results indicated that compared with the normal group,the content of GSH-Px in serums of the mice in group Ⅱ,Ⅲ,Ⅳ and Ⅴ significantly decreased( P < 0. 05); the differences in CAT content in each group were not significant( P > 0. 05);IL-4,IFN-γ and IL-10 content in serum of mice in group Ⅱ,Ⅲ,Ⅳ,and Ⅴ have difference( P < 0. 05). Compared with the model group,the amount of MDA in serums of mice in groupⅠ,Ⅲ,Ⅳ and Ⅴ significantly decreased( P < 0. 05); the SOD levels in serums of mice in groupⅠand Ⅴ rose significantly( P < 0. 05),the SOD levels in serums of mice in group Ⅲ and Ⅳ had a tendency of increasing( P > 0. 05); the content of IL-2 in serums of the mice in group Ⅲ,Ⅳand Ⅴsignificantly decreased( P < 0. 05); the differences of IL-4/IFN-γ ratios between groupⅠ,Ⅲ,Ⅳand Ⅴ increased significantly( P < 0. 05). Additionally,the Dendrobium officinale could adjust the serum antioxidant enzyme activity,Th1 and Th2 cells through controlling the cytokines of the mice whose immune systems are suppressed by CY,and it helps the blood cell indexes recover to a normal level. In conclusion,the Dendrobium officinale is able to effectively improve function of mice's immune systems which are suppressed by CY.
