枸杞多糖改善去卵巢大鼠脂质代谢紊乱的作用研究 被引量:11

Research on the Improvement Effects of LBP on Lipid Metabolism Disorders in Ovariectomized Rats

摘要 目的通过观察枸杞多糖对去卵巢大鼠血脂、肝脏形态及肝脏中P-GSK-3β和PPARγ蛋白表达的影响,探究枸杞多糖对去卵巢大鼠脂代谢的干预作用及机制。方法 30只SPF级健康雌性SD大鼠随机分为假手术组、去卵巢组、补佳乐组、枸杞多糖高剂量组及枸杞多糖低剂量组(每组6只)。全自动生化分析仪检测大鼠血脂水平,HE染色检测各组大鼠肝脏组织形态学变化,Western Blot方法检测肝脏中P-GSK-3β和PPARγ蛋白表达。结果血脂检测发现,与假手术组相比,去卵巢组TC、LDL-C水平显著升高(P<0.01),HDL-C水平显著降低(P<0.01),与去卵巢组相比,补佳乐组与枸杞多糖高剂量组TC、LDL-C水平显著降低(P<0.01),枸杞多糖低剂量组LDL-C水平降低(P<0.05)。HE染色发现,去卵巢组可见散在的脂肪空泡,枸杞多糖高剂量组脂肪空泡减少。Western Blot结果显示,与假手术组相比,去卵巢组P-GSK-3β与PPARγ表达水平显著下调(P<0.01);与去卵巢组相比,补佳乐组与枸杞多糖高剂量组P-GSK-3β与PPARγ表达水平升高(P<0.05或P<0.01)。结论高剂量枸杞多糖可能通过上调肝脏中p-GSK-3β与PPARγ表达改善脂代谢紊乱。 Objective To explore the effect of LBP on lipid metabolism in ovariectomized rats through observing the blood lipid, liver histolomorph and the expression of P -GSK -313 and PPART protein in liver. Methods 30 SPF healthy female SD rats were randomly divided into sham operation group, ovariectomized group, Progynova group, LBP high dose group and LBP low dose group. Blood lipid level were detected by fully automatic biochemical analyser. Liver tissue morphology were detected by HE stai- ning. The expression of P - GSK - 313 and PPAR~/ protein in liver were detected by Western Blot. Results Compared with the sham operation group, TC and LDL - C were significantly increased and HDL - C were significantly decreased in ovariectomized group (P 〈0.01 ) ; Compared with ovariectomized group, the level of TC and LDL- C in Progynova group and LBP high dose group were significantly decreased ( P 〈 0.01 or P 〈 0.05 ) . HE staining found that there are fat cavitations in ovariectomized group livers, while fat cavitations in LBP high dose group were significantly decreased. According to the results of western blot, compared with the sham operation group, the expression of P -GSK -313 and PPARγ protein in ovariectomized group were signifi- cantly increased (P 〈 0.01 ) ; Compared with ovariectomized group, the expression of P- GSK- 3βand PPART protein in LBP high dose group were significantly increased ( P 〈 0. 01 or P 〈 0.05 ). Conclusion High dose LBP may improve the lipid metabo-lism disorders through increasing the expression of P - GSK - 3 β and PPARγ protein of livers in ovariectomized rats.
