超高液相色谱法同时测定北五味子中7种木脂素类化合物 被引量:7

Simultaneous Determination of Seven Lignans from Schisandrae Chinensis Fructus by Ultra-performance Liquid Chromatography

摘要 建立了超高效液相色谱法(UPLC)同时测定北五味子中7种主要木脂素类成分的检测方法。采用ACQUITY UPLC~BEH C_(18)色谱柱(50 mm×2.1 mm,1.7μm);以水-乙腈为流动相,梯度洗脱分离,PDA检测波长254 nm,流速0.4 m L/min,柱温30℃,进样量2μL。优化了色谱分离条件,并进行了线性方程、准确度、重复性、回收率等方法学考察,在优化条件下,7种木脂素的出峰时间在5 min内,且线性关系良好(r>0.9994),方法检出限范围为0.0105~0.1600μg/m L,定量限为0.1050~0.8200μg/m L,相对标准偏差(RSD)在0.0340%~1.3800%之间,样品加样回收率在95.09%~104.50%范围内。本方法大大缩短了分析时间,灵敏度高,适用于北五味子中7种木脂素类成分的同时测定。 In this study, an uhra-performance liquid chromatography(UPLC) was developed for the simultaneous determi- nation of 7 lignans from Schisandrachinensis fruits. The analytes were separated on an ACQUITY UPLC BEH C18 col- umn(50 mm×2.1 mm,1.7μm) with acetonitrile-water as mobile phases in gradient elution under 254 nm. The flow rate was 0.4 mL/min and column temperature was set as 30 ℃. The injection volume was 2 μL. By using the optimized method, the detection limits of the 7 lignans ranged from0. 0105 to 0. 1600μg/mL, the limits of quantitation ranged from 0. 1050 to 0. 8200 μg/mL. The recoveries of the 7 lignans in the samples were 95.09% to 104.5% with relative stand- ard deviation( RSD% ) ranged from 0.0340% to 1. 3800%. The established UPLC method for the simultaneous determi- nation of 7 lignans can provide a more comprehensive reference for the quality control of S. chinensi.
