混合菌群FF降解染料效果及生物毒性变化 被引量:4

On the effects and biotoxicity variations as a result of dye biodegradation by bacterial consortium FF

摘要 利用混合菌群FF降解偶氮染料活性黑5和蒽醌染料活性蓝19,通过检测一些常规指标,如脱色率、DOC(溶解性有机碳)去除率、UV254、比紫外吸收值SUVA(UV254/DOC)等来表征降解效果,进一步通过固相萃取方法进行前处理,采用SOS/umu(又称umu试验)和双杂交酵母法分别检测染料降解过程中遗传毒性和抗雌激素活性。结果表明,采用混合菌群FF,活性黑5和活性蓝19(150 mg/L)在35℃和p H=8.0条件下静置培养48 h后脱色率分别为82.5%和89.2%;DOC去除率低,分别为19.5%和13.0%;降解后UV254和SUVA均增加。遗传毒性检测表明,活性黑5本身无遗传毒性,降解过程中也无遗传毒性;活性蓝19本身无遗传毒性,降解24 h、48 h后在高浓缩倍数下出现遗传毒性。随着降解的进行,活性黑5和活性蓝19的抗雌激素活性均显著增加。 This paper takes it as a study topic to degrade an azo dye(Reactive Black 5) and an anthraquinone dye(Reactive Blue 19) by bacterial consortium FF.To determine the treatment effects by the said bacterial consortium,we have made efforts in detecting some conventional indicators,such as,the decolourization ratio,DOC(dissolved organic carbon) removal ratio,UV254,SUVA(specific UV absorbance,as well as UV254/DOC).In so doing,we have pretreated and obtained the necessary samples in the dye biodegradation process with the solidphase extraction method.And,then,we have worked out the genotoxicity effects and the antiestrogenic activity of the samples by means of a SOS/umu assay and a yeast two-hybrid assay,respectively.And,we have successfully inoculated and cultured bacterial consortium FF with Reactive Black 5 and Reactive Blue19,respectively,on the condition of temperature 35 ℃,with the initial pH = 8.0,the initial mass concentrations of the above-said two dyes both being equal to 150 mg/L for the static culture.The results of our experimental studies indicate that we have been successful in increasing the decolourization effects of Reactive Black5 and Reactive Blue 19 under the static culture for 48 h,which can potentially be made to increase to 82.5% and 89.2%,respectively.On the other hand,DOC removal ratios are only19.5% and 13.0% through a 48 h-degradation.It is possible to increase the values of UV254 and SUVA by degrading Reactive black 5 and Reactive blue 19.In addition,the genotoxicity study reveals that no genotoxicity can be found before and after the biotreatment of Reactive Black 5 by bacterial consortium FF.And,no genotoxicity of Reactive Blue 19 has been found before the biodegradation,though it proves true to find the genotoxicity as a result of 24 h and 48 h biodegradation under the high concentrated multiplication.Finally,it can be concluded that with the help of the biodegradation process of Reactive Black 5 and Reactive Blue 19,the antiestrogenic activities of the above-mentioned two dyes have been increased significantly.
