摘要 目的:通过对银杏叶提取物黄酮类特征图谱和总黄酮醇苷含量的分析,对使用AB-8大孔树脂制备银杏叶提取物的工艺进行优化,得出最佳工艺。方法:用银杏叶对照提取物建立黄酮类化合物的特征图谱,以此为标准图谱,将制备的提取物与标准图谱中的17个特征峰比较,同时结合银杏叶对照提取物总黄酮醇苷的含量,综合评价银杏叶提取物的质量。结果:特征峰得分最高的为10号样品,得分最低的为5号样品;总黄酮醇苷含量得分最高的为9号样品和10号样品,得分最低的为2号样品;综合评价得分最高的为10号样品。结论:AB-8大孔树脂制备银杏叶提取物的最佳工艺为药液p H值5.0,先用25%乙醇洗脱除杂,再用75%乙醇洗脱。基于黄酮类特征图谱和总黄酮醇苷含量综合评分法可用于评价银杏叶提取物的质量,为提取物质量的评价方式提供一个新的思路。 Objective: Through the analysis of flavonoids characteristic chromatogram and the total flavonid content in the ginkgo biloba extract, to optimize the preparation of ginkgo biloba extract process by AB - 8 macroporous resin and conclud the best process. Method: Set up the preparation of characteristic chromatogram with ginko biloba reference sub- stance extract. Based on it,compare 17 characteristic peaks of the prepared extracts with the standard and combine with ginkgo biloba reference substance extract flavonoids content to evaluate the quality of ginkgo biloba extract. Results : The highest score of the characteristic peak was the sample No. 10 and the lowest score was the sample No. 5. And the highest content of total flavonoids was the sample No. 9 and No. 10 while the lowest score was 2 and comprehensive evaluation of the highest score was the sample No. 10. Conclusion: It is the best process for the Ginkgo biloba extract by AB -8 macro- porous resin of that when the liquor pH 5.0, first with 25% ethanol to wash and remove impurities, and then 75% etha- nol elution. Based on flavonoids characteristic chromatogram and total flavonoid content,we can evaluate the quality of the ginkgo biloba extract and it provides a new way of thinking for the evaluation of the quality of the extract.
