摘要
目的:观察不同刺激量艾灸关元穴对寒凝证类痛经大鼠扭体反应及脊髓内μ受体表达的影响,探讨不同灸量的镇痛效应及其部分中枢机制。方法:按随机数字表法将60只雌性SD大鼠,随机分为盐水组、寒凝证类痛经模型组、A组(艾灸1壮)、B组(艾灸3壮),每组15只。除盐水组外,其它各组采用全身冷冻法结合苯甲酸雌二醇注射液加缩宫素注射法制备寒凝证类痛经大鼠模型,A组和B组以不同灸量艾灸关元穴。观察大鼠扭体反应,并以荧光定量PCR(RT-PCR)法测定脊髓内μ受体mRNA的表达水平。结果:(1)扭体反应:与盐水组比较,模型组、B组扭体潜伏期缩短、扭体次数、扭体评分明显增高(P<0.05)。与模型组比较,A组、B组扭体次数、扭体评分明显减少(P<0.05)。(2)脊髓μ受体mRNA的表达:与盐水组比较,A组与B组的表达均增强(P<0.01),与模型组相比,A组与B组的表达增强(P<0.01)结论:(1)不同艾灸量施予关元穴对寒凝证类痛经大鼠模型均有镇痛作用。(2)艾灸产生镇痛效应的部分机制可能通过增强寒凝证类痛经模型大鼠脊髓内阿片受体的表达产生镇痛效应。
Objective : To investigate the analgesic effect and partly mechanism of the dysmenorrhea rats with cold stagnation syndrome by different moxibustion stimulus on CV4 by observing the writhing reaction and the μopioid receptor expression in spinal cord. Methods:Sixty female SD rats were divided into four groups according to the random number( n = 15 ) , including saline group, cold stagnation of primary dysmenorrhea model group, A stimulus group (1 cone) , B stimulus group ( 3 cones). Other groups except saline group preparation of cold stagnation of primary dysmenorrhea model through combining systemic freezing and injection estradiol benzoate and oxytocin, different moxibustion stimulus on A stimulus group and B stimulus group. Observe the writhing reaction and the μopioid receptor expression in spinal cord by RT- PCR. Results:①Writhing reaction:The writhing latency decreased significantly, the writhing times and the writhing scores per minute increased(P 〈 0. 05 ) of the model group and B stimulus group compared with that of the saline group. Com- pared with the model group, the writhing times and the writhing scores per minute obviously decreased( P 〈 0.05 ) of the A stimulus group and B stimulus group. ②The μopioid receptor mRNA expression in spinal cord:Compared with saline group ,the expression of A stimulus group and B stimulus group increased( P 〈 0. 01 ). Compared with model group, the expression of A stimulus group and B stimulus group increased( P 〈 0. 01 ). Conclusion: Dift^rent moxibustion stimulus on CV4 has analgesic effect on the dysmenorrheal rats with cold stagnation syndrome. The moxibustion can significantly in- crease the spinal cord ofp^opioid receptor expression of dysmenorrhea rats with cold syndrome.
