摘要
目的:探讨隔附饼灸对肠易激综合征大鼠脑肠互动轴的干预机理。方法:本实验参照AL-chaer E D等的方法制备IBS-D大鼠模型,随机分成正常组、模型组、隔药饼灸组和匹维溴铵组,每组8只。采用不同处理方法后,进行AWR评分,检测各组结肠、脊髓和下丘脑的orexin蛋白的表达。结果:与正常组比较,模型组AWR评分明显升高,差异存在统计学意义(均P<0.01),与模型组相比,隔药饼灸组和匹维溴铵组AWR评分明显降低,差异存在统计学意义(均P<0.01);与正常组比较,模型组大鼠的结肠、下丘脑、脊髓Orexin A阳性目标总面积和积分光密度表达均明显降低,差异存在着统计学意义(均P<0.05),与模型组比较,隔药饼灸组、匹维溴铵组大鼠的结肠、下丘脑、脊髓Orexin A阳性目标总面积和积分光密度表达表达明显增强,差异存在着统计学意义(均P<0.05)。结论:隔药饼灸治疗IBS的机理可能是刺激T10-13的神经节段,经脑肠互动轴调节CNS系统、ENS系统增加orexin蛋白的表达,降低内脏高敏感而提高大鼠的痛阈。
06jective: To explore the mechanisms of anconite cake - separated moxibustion on brain - gut interaction axis in rats with irritable bowel syndrome. Methods : Thirty - six female rats except 8 rats as control group were randomly selected to establish IBS model by modified method of AL - chaer E D. Twenty - four models rats were randomly divided into model group, anconite cake - separated moxibustion group and pinaverium bromide group, each group had 8 rats. After different processing methods were administrated, AWR scores were measured and orexin expressions in colon, spi- nal cord and hypothalamic by immunohistochemistry were detected. Results: Compared with normal group, AWR scores in model group under all kinds of pressures were significantly increased ( all P 〈 0.01 ). However, compared with model group, AWR scores of anconite cake - separated moxibustion group and pinaverium bromide group under all kinds of pressure were significantly decreased ( all P 〈 0.01 ). Compared with normal group, expressions of orexin in colon, spi- nal cord and hypothalamus were decreased. The differences between 2 groups were statistically significant ( all P 〈 0. 01). Compared with model group, expressions of orexin in colon, spinal cord and hypothalamus were increased. The differences between 2 groups were statistically significant ( all P 〈 0.01 ) Conclusions : Mechanism of anconite cake - separated moxibustion for treating IBS is probably to increase the expression of orexin and raise the pain threshold by ad- justing CNS system and the ENS system through brain - gut interaction axis to decrease visceral hyposensitivity and raise pain threshold .
