补肾活血法对膀胱出口部分梗阻大鼠膀胱逼尿肌组织氧化应激水平的影响 被引量:3

摘要 目的:观察膀胱出口梗阻(partial bladder outlet obstruction,PBOO)大鼠膀胱功能及逼尿肌组织氧化应激水平的变化,以及补肾活血方对这些变化的影响。方法:将3月龄雄性Wistar大鼠68只,随机选出12只大鼠作为正常组。剩余大鼠按后述手术方法复制膀胱出口梗阻(BOO)模型,并随机分为4组,每组14只,分别为模型组、补肾活血组、氯化铯组、补肾活血加氯化铯组。于造模成功并分组后第1天起,分别给予相应药物:生理盐水灌胃、补肾活血方灌胃、生理盐水灌胃+皮下注射氯化铯、补肾活血方灌胃+皮下注射氯化铯,正常组同期等体积生理盐水灌胃。给药30 d后,对各组大鼠进行膀胱测压;然后处死各组大鼠,快速取出膀胱并称取湿重;取逼尿肌组织用于检测MDA、SOD、T-AOC等氧化应激各项指标。结果:(1)与正常组相比,模型组(P<0.01)、氯化铯组(P<0.01)膀胱壁组织中MAD水平明显升高,中药组(P<0.01)及中药加氯化铯组(P<0.01)膀胱壁组织中MAD水平则明显低于模型组。(2)模型组(P<0.01)、氯化铯组(P<0.05)及中药加氯化铯组(P<0.01)膀胱壁组织中SOD水平亦明显高于正常组。中药组(P<0.01)膀胱壁组织中SOD水平则明显低于模型组。(3)模型组T-AOC水平明显低于正常组(P<0.05),其它各组与正常组相比,其膀胱壁组织T-AOC水平虽然均有所下降,但均无统计学意义。结论:PBOO可以引起膀胱组织总抗氧化能力下降,并最终导致氧化应激水平上升。补肾活血方可以明显降低大鼠膀胱组织MAD水平,而其对T-AOC的影响似乎并不显著。氯化铯可以部分阻断补肾活血方降低膀胱组织氧化应激水平的作用。 Objective : We try to observe the changes of bladder function and detrusor tissue oxidative stress level in the model rats of partial bladder outlet obstruction (PBOO). Meanwhile we observed the impact of Bushen Huoxue Formula on the changes. Methods : Firstly, 12 rats were randomly selected from 68 3 - month - old male Wistar rats as the normal group. And then the remaining rats were established BOO model according to the following surgical method. And the rats were randomly divided into four groups:model group, Bushen Huoxue Formula group, cesium chloride group, Bushen Huoxue Formula plus cesium chloride group, 14 rats in each group. The first day after established successfully and grouped, they were respectively given drug intervention: normal saline by lavage, Bushen Huoxue Formula by lavage, normal saline by lavage and cesium chloride by hypodermic injection, Bushen Huoxue Formula by lavage and cesium chloride by hypodermic injection. At the same time, the control group' s rats were given nomml saline by lavage. After 30 days, measure bladder pressure of each group. And then remove the bladder quickly and record the weights when all of rats were sacrificed. Take detrusor tissue to detect the oxidative stress indicators such as MDA, SOD, T - AOC. Results : (1)Compared with the control group, MDA levels in the bladder wall tissue of the model group ( P 〈 0. 01 ) and cesium chloride group (P 〈 0. 01 ) increased significantly; while Bushen Huoxue Formula group( P 〈 0. 01 ) and Bushen Huoxue Formula plus cesium chloride group(P 〈0. 01 ) were obviously lower than the model group' s. (2)SOD levels in the blad- der wall tissue of the model group (P 〈0. 01 ) , the cesium chloride group (P 〈0. 05) and Bushen Huoxue Formula plus cesium chloride group ( P 〈 0. 01 ) were higher than the control group' s. Bushen Huoxue Formula group was lower than the model group' s (P 〈0. 01 ). (3)T - AOC levels in the bladder wall tissue in the model group was obviously lower than the control group' s (P 〈 0. 05). Other groups' were lower than the control group' s. However, there was no statistical significance. Conclusion : The T - AOC in bladder tissue declines caused by PBOO and eventually makes oxidative stress levels rise. Bushen Huoxue Formula can significantly reduce MAD levels in bladder tissue, but its influence on T - AOC does not seem to be significant. That Bushen Huoxue Formula reduce oxidative stress level in the bladder tissue can be partially blocked.
