人参组培过程中SERK和WUS基因的表达特性

摘要 为研究人参叶片、愈伤组织以及从愈伤组织诱导出来的芽中与体细胞胚发生相关的基因的表达特性,在已报道的与植物愈伤组织诱导及体细胞胚发育密切相关的2个标志基因SERK和WUS,以actin为内参基因,采用RT-PCR方法分析这2个基因在人参叶片、愈伤组织及从愈伤组织长出来的芽中的表达特性。结果表明:虽然SERK和WUS在所有样品中均有表达,但在叶片和愈伤组织中的表达量无明显差异,而愈伤组织中分化出来的芽当中的表达量明显高于叶片和愈伤组织。 In order to elucidate expression patterns of the genes related with somatic embryogenesis in leaf, callus and callus shoot of Panax ginseng, two genes of SERK (SOMATIC EMBRYOGENESIS RECEP- TOR KINASE) and WUS (WUSCHEL) were selected from the genes which closely related with callus formation and somatic embryogenesis reported. And, actin gene as internal expression control standard, the expression properties of SERK and WUS genes in leaf, callus and callus shoot of Panax ginseng were detected by RT-PCR. The results showed that SERK and WUS were expressed in all the samples. The expression levels of both genes between leaf and callus were not significantly different, but their expres- sion levels in callus shoot were obviously higher than in leaf and callus.
