microRNA-126在多能干细胞定向分化为视网膜神经干细胞过程中表达量的变化及意义

Changes and Significances of MicroRNA - 126 Expression in Directional Differentiation of Pluripotent Stem Cells to Retinal Neural Stem Cells

摘要 目的研究microRNA-126(miR-126)在湿性老年黄斑变性(wetAMD)疾病患者来源的多能干细胞(iPSC)定向分化为视网膜神经干细胞(RNSC)中表达量的变化及意义。方法选取新发现的wetAMD患者10人作为观察组,无眼底病变及全身性疾病的10人作为对照组。抽取受试者的外周静脉血,提取单个核细胞(PBMC),进一步提取CD34’细胞,行腺病毒转染,诱导产生iPSC且进行鉴定;应用实时定量PCR(qRT—PCR)技术检测miR-126在10个不同诱导发育阶段的RNSC中的相对表达量。结果wetAMD疾病患者的外周血成功诱导分化出RNSC;观察组miR-126的不同诱导时间相对表达量的差异除时刻8、9、10外的测试时间点均有统计学意义(P〈0.05);对照组miR-126的不同诱导时间相对表达量的差异无统计学意义(P〉0.05);观察组与对照组细胞miR-126相对表达量在不同诱导时间的差异有统计学意义(P〈0.05)。结论wetAMD患者早期miR-126的表达量显著升高,且有逐渐降低的趋势。 Objective To investigate changes and significances of microRNA -126( miR- 126 )expression in directional differentiation of pluripotent stem cells (iPSCs)to retinal neural stem ceils (RNSCs)in patients with wet age -related macular degeneration(wet AMD). Methods Ten patients with wet AMD were chosen as observation group and ten healthy people were chosen as control group. Peripheral venous blood samples were collected, from which peripheral blood mononuclear cells (PBMCs)were extracted, and CD34^+ cells were extracted. CD34^+ ceils were transfected with adenovirus and iPSC was detected. Relative expression of miR - 126 at 10 different times in the process of RNSC were examined by quantitative real - time PCR( qRT - PCR). Results RNSC could be differentiated from pe- ripheral venous blood in patients with wet AMD. There were significant differences on miR - 126 expression at different times except time 8,9,10 in observation group ( P 〈 0.05 ) ; There were no significant differences on miR - 126 expression at different times in control group( P 〉 0.05 ) ;There were significant differences on miR -126 expression at different times between the two groups (P 〈 0.05 ). Conclusion The expressions of miR - 126 increase in patients with wet AMD and gradually decrease with time.
