葛根素调节成骨细胞体外增殖及相关信号通路表达改变的实验研究 被引量:2

摘要 目的:研究葛根素对成骨细胞体外增殖及相关信号通路表达的调节作用。方法:培养小鼠原成骨细胞MC353-E1,分为用10μg/mL葛根素处理的葛根素组以及不含药物和血清培养基处理的对照组,处理前及处理后12、24、48h时,采用MTS试剂盒测定细胞的增殖活力,采用酶联免疫吸附试剂盒测定PI3K/AKT/FoxO信号通路分子及凋亡基因的表达量。结果:处理后12、24、48h时,葛根素组的细胞活力均显著高于对照组,p-PI3K、p-AKT、p-FoxO1、p-FoxO3a的表达量均显著高于对照组,Bmi、Bax、CytC、AIF、Caspase-3的表达量均显著低于对照组;葛根素对细胞活力以及p-PI3K、p-AKT、pFoxO1、p-FoxO3a、Bmi、Bax、CytC、AIF、Caspase-3表达量的调控作用具有时间依赖性。结论:葛根素能够通过PI3K/AKT/FoxO信号通路促进成骨细胞的增殖。 Objective:To study the regulating effect of puerarin on osteoblast proliferation in vitro and related signal pathway expression.Methods:Mouse osteoblasts MC353-E1 were cultured and divided into the puerarin group that were treated with 10μg/mL puerarin and the control group that were treated with medium without drugs or serum.Before treatment as well as 12 h,24hand 48 hafter treatment,MTS kits were used to determine the cell proliferation activity,and enzyme-linked immunosorbent assay kits were used to determine the expression of PI3K/AKT/FoxO signal pathway molecules and apoptosis genes.Results:On 12 h,24hand 48 hafter treatment,the cell viability of puerarin group were significantly higher than those of control group,and p-PI3 K,p-AKT,p-FoxO1 and p-FoxO3 aexpression were significantly higher than those of control group while Bmi,Bax,CytC,AIF and Caspase-3expression were significantly lower than those of control group;puerarin had time-dependent regulating effect on the cell viability as well as p-PI3 K,p-AKT,p-FoxO1,p-FoxO3 a,Bmi,Bax,CytC,AIF and Caspase-3expression.Conclusions:Puerarin can promote osteoblast proliferation through PI3K/AKT/FoxO signal pathways.
