摘要
目的:探讨异丙酚对线粒体介导的乳大鼠心肌细胞氧化应激通路的作用。方法:取新生大鼠乳鼠(3~4 d)心肌细胞作为研究对象,实验分为对照组(C组)、H_2O_2组(终浓度500μM,H组)、异丙酚组(50μM异丙酚,P组)、同时使用H_2O_2和异丙酚组(500μM H_2O_2+50μM异丙酚,PH组)。采用流式细胞分析仪测定异丙酚对大鼠乳鼠心肌细胞ROS水平的影响,使用ELISA法测定各组心肌细胞线粒体途径介导的凋亡因子Caspase 3和Caspase 9活性变化。结果 :异丙酚对H_2O_2介导的大鼠乳鼠心肌细胞ROS水平增加具有明显的抑制作用(P<0.05)。异丙酚可明显下调线粒体途径介导的凋亡因子Caspase 9和Caspase 3的活性(P<0.01)。结论:异丙酚通过抑制心肌细胞ROS水平以及下调线粒体介导的凋亡因子Caspase 9和Caspase 3的活性发挥对心肌细胞的保护作用。
Objective: This study aimed to investigate the effects of propofol on ROS and apoptosis of neonatal rat cardiomyocytes (NRCMs). Methods: Cultured neonatal rat (3 ~ 4 days) cardiomyocytes were used in the study. The experiment included four groups: control group(Group C), H2O2 group(final concentration of 500 μM, Group H), propofol group(50 μM propofol, group P), and H2O2 and propofol group(500 μM H2O2 + 50 μM propofol, group PH). Flow cytometry(FCM) was used to measure the ROS level of NRCMs, and the activity of Caspase 3 and Caspase 9 were evaluated with ELISA. Results: Propofol significantly inhibited the ROS production (P 〈 0.05) and the activity of Caspase 3 and 9 (P 〈 0.01) induced by H2O2(P 〈 0.01) in neonatal rat cardiomyocytes. Conclusion: Propofol inhibited the ROS production and the activity of caspase 3 and caspase 9 of mitochondria, which may contribute to the protective effect of propofol on cardiomyocytes.
