摘要
建立了超声波辅助提取结合高效液相色谱荧光检测技术(HPLC-FLD)测定枸杞中4种生育酚的含量,考察了影响该前处理方法的因素包括萃取剂种类、料液比、温度、超声功率和萃取时间。结果表明,4种生育酚在质量浓度0.5~1 000μg/L之间线性良好,相关系数(r)均大于0.99,日内和日间重现性分别为4.6%和10.3%。并将此法成功应用于4种市售枸杞中生育酚含量的测定,平均加标回收率为83.2%~106.0%,相对标准偏差为1.54%~9.12%。
A convenient and reliable method was developed for the determination of four tocopherols(α-T, (β+γ)-T and δ-T) in lycium barbarum by using ultrasound extraction combined with HPLC-FLD. The parameters such as type of extraction solvents, solid-liquid ratio, temperature and ultrasound power and ex-traction treatment time were examined and optimized. The result showed that the linearity over a range of 0.5 -1 000 μg/L with correlation coeffcient of r 〉0.99. Intra-day and inter-day repeatability values ex-pressed as relative standard deviation were 4.6% and 10. 3%, respectively. The developed method was resoundingly applied to the determination of four tocopherols in lycium barbarum collected locally. Aver-age recovery varied from 83.2% to 106. 0% ,with RSD in the range of 1.54% to 9. 12% .
