Analysis of human papillomavirus specific cytotoxic lymphocyte immune response in head and neck carcinoma patients

摘要 手术、放疗、化疗被称之为肿瘤三大传统治疗手段。但是,近年来,肿瘤免疫疗法取得了令人瞩目的临床效果,成为目前最有潜力治愈恶性肿瘤的手段。尤其与T细胞免疫相关的免疫检查点阻滞剂(如,抗CTL-A4单抗、抗PD-1单抗),快速获得美国FDA的批准,应用于恶性黑色素瘤、非小细胞肺癌、头颈肿瘤、尿路上皮癌、霍奇金淋巴瘤的临床实践;同时在肝癌、胃癌、结直肠癌和其他肿瘤也显示出了可喜的前景。随着人们对机体抗肿瘤免疫应答及肿瘤免疫逃逸机制的深入认识, Objective To provide experimental evidence for immunotherapy by investigating the level of specific cells immune responses in patients with head and neck carcinoma. Methods 55 cases (the control group, Head and neck cancer group) were newly diagnosed HNC patients and 100 cases healthy (the con-trast group) , and all patients were pathological diagnosis confirmed. Specific immune response level of pe-ripheral T cells in patients and healthy to El, E2 , E4, E5 , E6 , E7 , LI and L2 entire protein antigen pep-tide pools of HPV genome sequences (including E region and L zone) expression were measured by using ELISPOT assay. Results The frequency of CTL immunoreactivity in E1-L2 peptide pool was significantly higher than that in the healthy control group (P 〈C0.05) ; The intensity of HPV E6 , E7 antigen peptide pools specific CTL immune reaction in HNC patients were significantly higher than the healthy group , which had statistically significant (P 〈C0.05). Conclusion HPV antigen E6 , E7 proteins may be oncogenic proteins leading to the occurrence and development of head and neck tumors, potentially ideal targets for head and neck cancer immunotherapy.
