天山岩黄芪总黄酮含量的测定及抗氧化活性研究 被引量:1

摘要 目的测定天山岩黄芪总黄酮含量及其抗氧化活性。方法采用回流提取法,料液比为1∶30,70%乙醇70℃回流3h,提取天山岩黄芪中总黄酮,并以紫外分光光度法测定其中总黄酮含量;以Vc为阳性对照,测定天山岩黄芪总黄酮对DPPH·(超氧阴离子)与·OH(羟基自由基)的清除作用。结果回归方程为Y=11.663 X+0.007 9,r=0.999 6,线性范围:19.3~67.6μg/mL;方法精密度良好,RSD=1.2%;方法重现性良好,RSD=2.0%;在0~75min内样品稳定性良好,RSD=1.1%;样品回收率为99.9%,RSD=1.6%。天山岩黄芪中总黄酮含量为11.25mg/g,RSD=1.2%。天山岩黄芪总黄酮对DPPH·与·OH的清除率有浓度依赖性,当总黄酮浓度为50μg/mL时,对DPPH·的清除率为59.821%,对·OH的清除率为25.782%。结论方法简单易行、准确可靠,适用于天山岩黄芪总黄酮的含量测定,天山岩黄芪总黄酮具有较好的抗氧化活性。 Objective To determine the total flavonoids content and antioxidant activity of Hedysarum semenovii Regl. et Herd. Methods The total flavonoids extracted from Hedysarum semenovii Regl. et Herd by refluxing extract method, material liquid ratio was 1 : 30 ,and we refluxed extraction by 70% al-cohol in the 70℃ for 3 h, and tested the content of total flavonoids by the UV; Additionally, we also tested the total flavonoids antioxidant activity by scavening rate of DPPH , (superoxide anion) and , OH (hydroxyl radi-cal) ,finally, we compared the results to the VC positive control group. Results The regression equation was Y =11.6 63X + 0.0079,r = 0.999 6, and linearity range was 19.3 ,67.6 μg/mL; and the sample had a good precision and repeatability, respectively, RSD=1.2%, 2.0% ;and the sample stability was good in 0-75 min,RSD =1.1%; besides, the recovery rate of the sample was 99.9%, RSD= 1.6%. Therefore, the con-tent of total flavonoids in Hedysarum semenovii Regl.et Herd was 11.25 mg/g, RSD=1.3%. The scaven-ging rate of total flavonoids in Hedysarum semenovii Regl. et Herd to DPPH , and , OH depended on concentration, the clearance rate to the DPPH , was 59.821 % when the total flavonoids concentration was 50 jug/mL, and in the same concentration, the total flavonoids clearance rate was 25.782% to the , OH. Conclusion This method is simple, accurate, and reliable, which can be used for the Hedysarum semenovii Regl.et Herd flavonoids content determination, and it has a good antioxidant activity.
